摘要
目的:国内与国外文献报道白术内酯Ⅱ作用于肿瘤细胞有效剂量差异较大,为探索其实际作用剂量以及出现差异的原因进行实验研究。方法:白术内酯Ⅱ20 mg以DMSO 400μL溶解,并以纯水进一步稀释白术内酯Ⅱ溶液至5、25、50、100、150、300μg/m L,利用紫外分光光度法检测白术内酯Ⅱ实际作用浓度。同法检测胎牛血清、双抗对实际浓度的影响。结果:紫外分光光度法检测上述配制浓度所对应的实际浓度分别为3.81、11.28、13.01、14.94、15.62、17.39μg/m L,并且得出胎牛血清、双抗对白术内酯Ⅱ实际浓度并无影响。结论:国内文献报道白术内酯Ⅱ作用于肿瘤细胞的实际有效浓度与国外文献报道的作用浓度相当,而国内文献报道实验中白术内酯Ⅱ的作用剂量较大的原因并非受培养基影响,而可能与药物溶解度及溶剂相关。
Objective: The domestic and foreign literatures reported different effective dose of AtractylenolideⅡ(AT-Ⅱ),and some experiments need we conduct to explore the actual effective dose of AT-Ⅱ and make sure the reasons for the differences.Methods: 20 mg of AT-Ⅱ was dissolved in 400 μL of DMSO to obtain a concentration of 50×103μg/m L.Further,the above-obtained solution of AT-Ⅱ was diluted to a concentration of5,25,50,100,150,300 μg/m L by water.Then the actual concentration of AT-Ⅱ was determined by ultraviolet spectrophotometry.The effect of fetal bovine serum and double antibody on the actual concentration was detected by the same method.Results: The actual concentration were 3.81,11.28,13.01,14.94,15.62,17.39 μg/m L,and there is no difference whether it contains the fetal bovine serum or double antibody.Conclusion: The effective dose of AT-Ⅱ reported in domestic literatures is larger,and the real concentration of AT-Ⅱ which is effective to the tumor cells reported in China is consistent with the concentration reported overseas.In addition,it is not related to the medium but the solubility of AT-II and the solvent.
