缺位型硅钨酸盐对HepG2细胞增殖的抑制作用

摘要 目的研究缺位型硅钨酸盐K_8[γ-SiW_(10)O_(36)]·12H_2O(简称KSiW_(10))的体外抗肿瘤活性。方法应用四甲基偶氮唑盐比色法分析KSiW_(10)对人肝癌细胞(HepG2)的增殖抑制活性。用流式细胞术分析细胞凋亡,计算各期细胞比例及细胞凋亡率。结果缺位型硅钨酸盐KSiW_(10)对HepG2细胞有增殖抑制作用,具有药物浓度依赖性,IC^(50)值为(9.7±3.6)×10^(-5)mol/L。不同浓度KSiW_(10)处理24 h凋亡细胞所占比例显著增加且呈剂量依赖性。结论硅钨酸盐KSiW_(10)能够抑制肿瘤细胞增殖,诱导细胞凋亡。 Objective To study the anti-tumor activity of lacunary silicotungstate 盐 K8[γ-SiW10 O(36)]·12 H2 O( abbreviated form: KSiW10) in vitro. Methods The inhibitory effect of KSiW10 on the growth of human hepatoma HepG2 cells was observed by MTT( 3-[4,5-dimethylthiazo-2-y]-2,5-diphenyltetrazolium bromide) assay. Flow cytometry was applied to analyze cell apoptosis by calculating the ratio of tumor cells in different phase and its apoptotic ratio. Results KSiW10 inhibited HepG2 cells proliferation in dose-dependent manner,with IC50 value of( 9. 7 ± 3. 6) × 10^-5 mol/L. After treating with different concentrations of KSiW10 for 24 h,the apoptosis proportions of tumor cells increased in dose-dependent. Conclusion KSiW10 could inhibit tumor cells growth and induce cell apoptosis.
