摘要
目的建立高效液相色谱法同时测定泽泻中23-乙酰泽泻醇B和24-乙酰泽泻醇A的含量。方法采用Diamonsil C_(18)(2)柱(250 mm×4.6 mm,5μm);流动相为乙腈-水(80∶20,v/v),流速为0.8 mL/min,检测波长为208 nm,柱温为35℃,进样量为20μL。结果 23-乙酰泽泻醇B和24-乙酰泽泻醇A均在1~50 mg/L(R=0.999 5)浓度范围内与峰面积呈良好的线性关系,生泽泻的平均回收率分别为99.6%和98.9%,盐泽泻的平均回收率分别为99.1%和99.8%。结论方法简便、快速、具有良好的重现性和稳定性,可用于泽泻药材的质量控制。
Objective To develop a high-performance liquid chromatography method for determination of the content of Alisol B-23-acetate and Alisol A-24-acetate in Rhizoma Alismati. Methods The analysis was performed on Diamonsil C18( 2) column( 250 mm × 4. 6 mm,5 μm) with isocratic elution( 80∶ 20,v/v) by using the mobile phase of acetonitrile( A) and water( B). The flow rate was 0. 8 mL/min. The detecting wavelength was set at 208 nm and the column temperature was maintained at 35 ℃. Results The linear ranges were 1 ~ 50 mg/L( R = 0. 999 5) for Alisol B-23-acetate and 1 ~ 50 mg/L( R = 0. 999 5) for Alisol A-24-acetate. The average recoveries in crude Rhizoma Alismatis were 98. 9% for Alisol B-23-acetate and 99. 6% for Alisol A-24-acetate. The average recoveries in salted Rhizoma Alismatis were 99. 1% for Alisol B-23-acetate and 99. 8% for Alisol A-24-acetate. Conclusion The method is simple,rapid,stable and reproducible,which could be used for the quality control of Rhizoma Alismatis.
