低强度激光修复术对小型巴马猪退变椎间盘水通道蛋白-3表达的影响 被引量:2

摘要 目的:通过构建小型巴马猪椎间盘退行性病变模型,观察低强度激光对猪椎间盘水通道蛋白-3(Aquaporin-3,AQP-3)表达的影响。方法:小型巴马猪9头,随机分为对照组(Con组),造模组(Mod组)及激光治疗组(Las组),每组3头。Con组不进行任何处理,Mod组及Las组在C形臂X光引导下利用16G针刺入猪L2-3、L3-4、L4-5、L5-6椎间盘进行破坏,Las组猪椎间盘退变模型建立1个月后,对已处理的椎间盘进行低能量激光治疗再观察1个月,进行MR检测,并取椎间盘HE染色、免疫组化及Western检测AQP-3的表达情况。结果:三组在造模前、造模后1个月、治疗后1个月,Con组猪的椎间盘T2加权像呈高信号,Mod组信号强度明显下降,Las治疗组呈中、高信号影。HE染色Con组结构完整,Mod组结构遭到破坏,Las组结构较Mod组显著恢复。与Con组比较,Mod组和Las组猪椎间盘的AQP-3表达均显著降低(P<0.05),而Las组AQP-3表达较Mod组显著升高,差异有统计学意义(P<0.05)。结论:低强度激光对小型巴马猪退变的椎间盘具有保护作用,其机制可能与上调其间盘组织水通道蛋白表达有关。 Objective: To investigate the effect of low intensity laser irradiation on porcine intervertebral disc aquaporin-3 (AQP-3) expression by constructing the miniature Bama pig degenerative disc model. Methods: Nine miniature Bama pigs were randomly divided into three groups: control group (Con), model group (Mod) and laser treatment group (Las). Con group did not undergo any treatment, while the Mod group and the Las group underwent Type C arm X-ray guidance puncture with 16G needles to break down the L2-3, L3-4, L4-5, L5-6 intervertebral discs. After one month, the discs were treated with low-energy laser treatment for another month. MR imaging was performed for 1 month. The expression of AQP-3 was detected by HE staining, immunohistochemistry and Western blot. Results: The T2-weighted images of the intervertebral discs showed a high signal intensity in the three groups before modeling, one month after modeling and one month after treatment. The signal intensity of the Mod group decreased significantly, and the signal intensity in the Las group was moderate and high compared with the Mod group. The structure of Con group was damaged and the structure of Las group was restored compared with Mod group. Compared with Con group, the expression of AQP-3 was significantly decreased in Mod and Las groups (P 〈 0.05), but the expression of AQP-3 in Las group was significantly higher than Mod group (P 〈 0.05). Conclusion: Low intensity laser has a protective effect on degenerated intervertebral disc of Bama miniature pig, and its mechanism is possibly related to the up-regulation of aquaporin expression.
