银杏内酯A、B联合改善缺血/再灌注损伤老年大鼠心功能 被引量:2

摘要 目的探讨银杏内酯A(GA)与银杏内酯B(GB)联合用药后处理对缺血/再灌注(I/R)损伤的老年大鼠心功能的影响。方法将老年SD大鼠随机分为正常对照组(Control组)、缺血/再灌注损伤组(I/R组)、GA后处理组(GA组)、GB后处理组(GB组)及GA、GB联合用药组(GA+GB组)各10只。通过对离体心脏进行停灌30 min再灌注60 min制备离体心脏I/R损伤模型,并记录左心室收缩压(LVSP),左心室舒张末压(LVEDP),左心室内压上升和下降的最大变化速度(±dp/dtmax),收集冠脉流出液检测其中的乳酸脱氢酶(LDH)含量。结果 GB与GA+GB无论在复灌30 min还是在复灌60 min都能明显地抑制缺血后心室内压差ΔLVP、+dp/dtmax和-dp/dtmax各指标的降低,差异有统计学意义(P<0.05)。与I/R组比较,GB与GA+GB处理均能够明显降低冠脉流出液中LDH的含量,差异有统计学意义(P<0.01)。结论GA与GB联合用药后处理能够改善缺血/再灌注损伤的离体老年大鼠的心功能。 Objective To explore the effects of ginkgolide A,ginkgolide B combined post-treatment therapy on the cardiac function of aged rats hearts with ischemia/reperfusion injury. Methods Sprague-Dawley aged rats were randomly divided into five groups: a normal control group( Control group), an ischemia/reperfusion injury group( I/R group),a ginkgolide A post-treatment group( GA group),a ginkgolide B posttreatment group( GB group) and a combined post-treatment group( GA+GB group). An in vitro isolated aged rat heart model of I/R injury was established by the whole heart stopped perfusion 30 min followed 60 min reperfusion.Then,the left ventricular systolic pressure( LVSP),left ventricular end diastolic pressure( LVEDP),the maximal rate in rise and fall of ventricular pressure( +dp/dtmax,-dp/dtmax) were recorded and the activity of LDH in the fluid of coronary artery were tested. Results Both GB and GA + GB could significantly inhibit the decrease of LVP,+dp/dtmaxand-dp/dtmaxindexes after ischemia,and the difference was statistically significant( P〈0.05)between 30 min and 60 min after reperfusion. Compared with the I/R group,GB and GA + GB treatment can significantly reduce the content of LDH in the coronary outflow fluid,the difference was statistically significant( P〈0.01).Conclusions Combined administration of GA and GB can improve the cardiac function in the aged rats with ischemia/reperfusion injury.
