麦芽酚铝对PC12细胞钙稳态和凋亡的影响 被引量:1

摘要 目的探讨麦芽酚铝[aluminum maltolate,Al(mal)_3]染毒对大鼠肾上腺嗜铬细胞瘤细胞(rat pheochromocytoma cells,PC12)钙稳态及凋亡的影响。方法将PC12细胞随机分为0(对照组)、50、100、200、400μmol/L Al(mal)_3染毒组,分别培养12、24、48 h,采用CCK-8试剂盒检测PC12细胞活性,以流式细胞术检测PC12细胞内游离钙离子荧光强度[Ca(~2+)]i及细胞凋亡率。结果随着Al(mal)_3染毒时间和染毒剂量的增加,PC12细胞活性逐渐下降,细胞[Ca(~2+)]i与细胞总凋亡率逐渐升高。除50μmol/L Al(mal)_3染毒12、24 h及100μmol/L Al(mal)_3染毒12 h外,其他各剂量组染毒PC12细胞12、24、48 h后细胞活性均低于对照组,细胞[Ca(~2+)]i均高于对照组,差异有统计学意义(P<0.05)。400μmol/L Al(mal)_3染毒PC12细胞12 h,200、400μmol/L Al(mal)_3染毒24 h,100、200、400μmol/L Al(mal)_3染毒48 h后,PC12细胞总凋亡率均高于对照组,差异有统计学意义(P<0.05)。结论钙超载可能是Al(mal)_3引起凋亡发生的主要机制。 Objective To explore the influence of Al(mal)3 on the intracellular Ca2+ homeostasis and apoptosis of PC12 cells. Methods The PCl2 cells were randomly divided into five groups, such as 0 (control group), 50, 100, 200, 400 μmol/L Al(mal)3 groups. After continuous cultivation for 12 h, 24 h, 48 h, cell activities were detected by using a cell counting kit 8 (CCK-8) assay, the intensity of intracellular Ca2+ fluorescence ([Ca21] and effect of Al(mal)3 on cell apoptosis rate were analyzed by using ~flow cytometry. Results The activities of PC12 cells were decreased gradually and the [Ca2~]~, the total apoptosis rate of PC12 ' cells were increased significantly with the increase of Al(mal)3 dosage and exposure time.The cell activities significant decreased, the [Ca2+] significantly increased in Al(mal)3 group exposed for 12 h, 24 h, 48 h compared with those in control group (P〈0.05), except 50μmol/L Al(mal)3 group exposed for 12 h, 24 h and 100 μmol/L Al(mal)3 group exposed for 12 h (P〉0.05). The total apoptosis rates in 400 μmol/L Al(mal)3 group exposed for 12 h, 200 μmol/L and 400 μmol/L Al(mal)3 groups : exposed for 24 h, 100μmol/L, 200μmol/L and 400μmol/L Al(mal)3 groups exposed for 48 h,were higher than those in control group(P〈0.05). Conclusion Calcium overload may be the main mechanism of apoptosis induced by Al(mal)3.
