摘要 以一株海洋Bacillus sp.细菌为对象,进行天然药物化学研究性实验的教学探索。通过优化菌株发酵条件、改进文献提取分离纯化方案,高效率地分离纯化出目标产物Iturin A,并指导学生综合运用质谱,1HNMR结合文献调研鉴定产物结构。该实验合理使用制备高效液相色谱仪,液质联用仪、核磁共振仪等大型仪器设备,改进了传统天然药物化学实验内容,实现了学生实验中微量天然产物快速、高效、可视化的分离与纯化。实验有助于激发学生学习兴趣,使其全面深刻地认识微生物天然产物的科学研究流程,了解掌握不同大型仪器的应用范围和操作技术,提升操作技能和创新能力。 A research experiment of natural medicinal chemistry was designed by using a Bacillus sp. which belongs to marine bacteria. The target was efficiently to isolate product Iturin A by optimizing fermentation conditions, improving the methods of extraction and separation presented in previous references. Moreover, under teachers' guidance the structures of the product were elucidated by the students using MS, 1HNMR and document investigation. In this experiment, the large-scale instruments including preparative HPLC spectrometer, LCMS spectrometer, and NMR spectrometer were reasonably used and the traditional teaching contents in natural medicinal chemical experiments werere formed. The fast, efficient and visual separation and purification of micro-scale natural products was achieved in the experiment. The experiment helps students to enhancethe interest of study, deepen the understanding of the scientific research procedures of marine microbial natural product chemistry, grasp the application scope and operation technology of these large-scale instruments, and improve the operating skill and innovation ability.
