穴位敷贴含药血清促进神经干细胞增殖作用研究 被引量:2

摘要 目的:探讨安宫牛黄贴含药血清对神经干细胞(neural stem cells,NSCs)增殖的影响以及对分泌型血管内皮生长因子(vascular endothelial growth factor,VEGF)表达的作用。方法:将未分化状态的C17.2细胞培养并传代,按含药血清10%给药,分为穴位敷贴高剂量组,穴位敷贴中剂量组,穴位敷贴低剂量组,药物组,以及空白组。MTT法检测NSCs增殖活性,ELISA法检测VEGF蛋白表达。结果:MTT数据显示,安宫牛黄贴穴位敷贴能有效促进NSCs的增殖,以穴位敷贴低剂量、中剂量组效果较为显著(P<0.05)。ELISA结果显示,穴位敷贴低剂量、中剂量、高剂量均能不同程度升高神经干细胞表达分泌型VEGF蛋白,其中穴位敷贴中剂量组增高分泌型VEGF蛋白含量的效果最好(P<0.05)。结论:VEGF分泌增多对体外培养的神经干细胞增殖有明显促进作用,穴位敷贴含药血清正是由于增加了神经干细胞表达分泌型VEGF蛋白,从而促进了神经干细胞的增殖。 Objective: To investigate the effect of proliferation of NSCs and expression of secreted VEGF by drug serum with point application of Angong Niuhuang Acupoints. Methods: Undifferentiated C17. 2 cells were cultured and passaged,then 10% drug serum were administered,including acupoint application group of high dose,middle dose and low dose,positive drug group,blank control group. MTT assay were used to detect NSCs proliferation,while ELISA were used to test VEGF expression. Results: MTT data showed that acupoint application can effectively promote the proliferation of NSCs,in which the effect of low and media dose group was significant( P〈0. 05). ELISA results suggest that acupoint application of low,media and high dose can all increased the expression of secreted VEGF in NSCs on different extent,in which the media-dose group has better effect( P〈0. 05). Conclusion:The secretion of VEGF can clearly promote the proliferation of NSCs,therefore the drug serum with acupoint application promoting the proliferation of NSCs by increasing the expression of secreted VEGF.
