雄激素合成酶在H22荷瘤小鼠不同证候表达差异 被引量:1

摘要 目的:研究睾丸雄激素合成酶与肿瘤状态下小鼠证候的关系。方法:采用实验小鼠及其标准化四诊与辨证方法筛选H22荷瘤小鼠邪毒壅盛证、气虚证、阳气虚证、气阴阳虚证,并以Gene Chip Mouse Exon 1.0 ST Array技术检测睾丸组织基因芯片表达谱,重点分析睾丸基因芯片中雄激素合成酶及其调节因子在不同证候小鼠中的差异表达。结果:与正常组比较,Star、Stard5、Cyp17a1、Hsd3b6、Hsd17b3、Sult1e1等基因在早期邪毒壅盛证和气虚证小鼠呈现下调趋势,且在中晚期阳气虚证和气阴阳虚证下调幅度更加显著,Cyp11a1、Hsd3b1、Hsd17b10、Hsd17b11、Hsd17b12、Fdx1、Fdxr、Por、Sult1a1等仅在中晚期阳气虚证和气阴阳虚证中显著下调,Cyp17a1仅在睾丸组织特异性中表达。结论:H22荷瘤小鼠肿瘤形成后,自发产生的虚证越严重,其睾丸雄激素合成酶基因转录活性越低。 Objective: To study the relation of androgen synthases genes expression and syndromes in H22 tumor Mice. Methods: By the quantitative four diagnosis and syndrome differentiation methods, different H22 tumor mice with poisonous pathogenic factors syndrome and asthenia of qi syndrome in earlier period, asthenia of yang and qi syndrome on intermediate stage, asthenia of qi and yin and yang syndrome on advanced stage were obtained. Then testis cDNA microarray were detected by GeneChip Mouse Exon 1.0 ST Array. The androgen synthase and its regulatory factor genes expression were analyzed. Results: Compared with normal mice, Star, StardS, Cypl7al, Hsd3b6, HsdlTb3 and Suhlel genes expression were down-regulated in pathogenic factors syndrome and asthenia of qi syndrome on earlier period, and those genes were more significantly down-regulated in asthenia of yang and qi syndrome, asthenia of qi and yin and yang syndrome. Cypllal, Hsd3bl, Hsdl7blO, Hsdl7bll, Hsdl7bl2, Fdxl, Fdxr, Por and Sultlal genes expression were down-regulated only in asthenia of yang and qi syndrome, asthenia of qi and yin and yang syndrome. And Cypl7al specifically expressed only in testis. Conclusion: After the tumorigenesis, androgen synthases genes expression were decreased with spontaneous asthenia svndromes aggravation.
