摘要 目的:采用正交设计法优化纤维素酶提取滁菊中黄酮类成分。方法:以木犀草素、香叶木素、芹菜素的含量为指标,高效液相色谱仪测定其含量,正交试验设计法优选纤维素酶提取滁菊中黄酮类成分工艺。结果:最佳提取工艺为温度45℃,pH 5.0,酶解1.5 h,酶用量0.6%,在此条件下,木犀草素、香叶木素、芹菜素的含量分别为1.49%、1.64%、1.69%。结论:在最佳酶解条件下,纤维素酶能显著地提高滁菊黄酮类成分的提取率。 Objective:To optimice the extraction of flavonoids from Dendranthema morifolium by Cellulase by or- thogonal design. Methods:The contents of luteolin, diosmetin, apigenin were measured. The content of fla- vonoids in Chrysanthemum morifolium was determined by orthogonal design. Results: The optimum extraction conditions were at temperature 45 ℃, pH 5.0, enzymatic hydrolysis 1.5 h, enzyme dosage 0. 6 %. Under these conditions, the content of luteolin, diosmetin and apigenin was 1.49 %, 1.64 % and 1.69 % respectively. Conclusion: Under the optimum conditions of enzymatic hydrolysis, the cellulase can significantly improve the extraction rate of active ingredients of Dendranthema morifolium.
