摘要
建立了一种测定鸡肉或者鸡蛋中环丙沙星、达氟沙星、恩诺沙星、沙拉沙星4种氟喹诺酮类药物残留量的高效液相色谱法。鸡肉或鸡蛋样品两次用磷酸盐缓冲溶液(p H=7.0)提取,正己烷去杂质,固相萃取用C18小柱净化,流动相调整为0.05 mol/L磷酸溶液(p H=2.4):乙腈比例为70:30,收集洗脱液,用高效液相色谱法测定。添加低、中、高浓度的标准品时,回收率为88.68%~100.30%,相对偏差在1.55%~2.31%(n=5);标准曲线显示环丙沙星、恩诺沙星和沙拉沙星在0.001~0.200 mg/kg,达氟沙星在0.001~0.040 mg/kg范围内线性关系良好,相关系数在0.9999以上。采用本方法操作简便,灵敏度高,专属性强,基质干扰小,结果准确可靠,可以满足鸡肉或者鸡蛋中的环丙沙星、达氟沙星、恩诺沙星、沙拉沙星四种氟喹诺酮类药物残留检测的需要。
The research aimed to develop a method for the determination of four kinds of fluoroquinolones,including ciprofloxacin,dafoxacin,enrofloxacin and salazacin in chicken or egg by high performance liquid chromatography(HPLC).Chicken or egg samples were extracted twice with phosphate buffer solution(pH = 7.0) and decolored with n-hexane.Using C18 solid phase extraction cartridges and ratio of mobile phase adjusted to be 0.05 mol/L phosphoric acid solution(pH = 2.4) during the elution.The ratio of acetonitrile to acetonitrile was70: 30.The eluate was collected and measured by high performance liquid chromatography.The recoveries were 88.68% ~ 100.30% and the correlation coefficients ranged from 1.55% to 2.31%(n = 5).The calibration curve showed that ciprofloxacin,enrofloxacin and salvofloxacin were in the range of 0.001 ~ 0.200 mg/kg and 0.001 ~ 0.200 mg/kg,respectively.The correlation coefficient was above 0.9 999.The method is simple,sensitive,specific,small matrix interference,accurate and reliable to meet the determination of ciprofloxacin,dafoxacin,enrofloxacin and salazacin residues in the chicken or egg.
