贞术消积汤抑制STAT3/Survivin信号通路诱导肝癌细胞自噬实验研究 被引量:12

Expenimental Study of Zhenzhu Xiaoji Decoction Inducing Autophagy via STAT3/Survivin Signal Pathway in Hepatoma Cells

摘要 目的:通过观察贞术消积汤作用H22移植瘤后细胞形态学变化,探讨该复方体内抗肿瘤的分子机制。方法:将小鼠肝癌细胞移植于ICR小鼠皮下,贞术消积汤灌胃给药10 d;采用苏木素-伊红(HE)染色,光镜下观察各组细胞形态的变化;应用透射电镜观察模型组和贞术消积汤组细胞超微结构的改变;荧光定量PCR法检测Survivin mRNA表达;免疫组化法检测信号转导与转录活化因子3(STAT3)蛋白和抗凋亡因子(Survivin)蛋白表达。结果:光镜下观察发现,用药组细胞排列稀疏,组织产生大量空泡;透射电镜观察可见,贞术消积汤组细胞内发现自噬体和自噬溶酶体;与模型对照组相比,贞术消积汤组Survivin mRNA低于模型组;贞术消积汤组STAT3和Survivin表达下调。结论:贞术消积汤体内抗肿瘤的作用机制可能与其诱导细胞自噬有关,并通过抑制STAT信号通路发挥作用。 Objective : To explore the in vivo antitumor mechanism of Zhenzhu Xiaoji decoction by detecting the morphology changes of H22 cells. Methods: Murine hepatocarcinoma cells were injected into ICR mice and then they were given Zhenzhu Xiaoji decoction once a day for ten continuous days. The changes of cell mor- phology were observed under light microscopy and transmission electron microscopy(TEM) ; the expression of Survivin mRNA was detected by RT - PCR; and the protein expressions of STAT3 and Survivin were observed by IHC. Results: Under light microscopy, cell lineage was sparse and lots of vacuoles were produced in tis- sues with the medication; under TEM, there were autophagosome and autolysosome in cells; the expression of Survivin, Survivin mRNA, and STAT3 were down- regulated in the medication group compared to the model control group. Conclusion: The in vivo antitumor effect of Zhenzhu Xiaoji decoction is probably related to in- ducing cell autophagy by inhibiting STAT3/Survivin signal pathway.
