摘要 对微生物学实验教材上介绍的Schaeffer-Fulton芽胞染色法进行了改良。复染剂用1∶10稀释石炭酸复红染液代替0.5%番红水溶液,并对比不同初染剂孔雀绿溶液浓度与初染时间对染色效果的影响。经过多次实验,结果表明,以7.6%孔雀绿溶液为初染液,初染5 min效果最佳,芽胞的鲜绿色与菌体的红色对比明显,背景干净,易于观察,学生更容易鉴别芽孢和菌体。此染色方法简单可行,效果良好,适用于一般教学条件的学校实验室进行枯草芽孢杆菌的染色。 In this paper, the Schaeffer-Fulton spore staining method introduced in the microbiology experimental teaching material was improved. The counter stain was replaced with 1:10 dilute carbolic acid complex red dye instead of 0.5% safranin solution, and the effect of different primary stain malachite green solution concentration and initial dyeing time was compared. After many experiments, the results showed that the best effect was obtained at 5 min after initial dyeing with 7.6% malachite green solution as the initial dye solution. The bright green of the spores was obviously compared with the red of the bacterial cells. The background was clean and easy to observe. Students could identify the spores and bacteria more easily. This staining method is simple, feasible, well-effected and suitable for general teaching conditions in the laboratory of Bacillus subtilis staining.
