疏血通注射液对急性脑梗死缺血再灌注氧化应激水平的影响 被引量:11

Effect of Shuxuetong on Oxidative Stress Levels in Rats with Acute Cerebral Infarction Ischemia/Reperfusion

摘要 目的探讨疏血通对急性脑梗死缺血再灌注损伤氧化应激水平的影响。方法 60只大脑中动脉闭塞后再灌注模型大鼠随机分为模型组、低剂量组和高剂量组,同时20只大鼠作为假手术组。低剂量组和高剂量组大鼠分别经灌胃给予疏血通50 mg/kg和100 mg/kg,模型组和假手术组经灌胃给予等体积PBS溶液。治疗48 h后测定4组大鼠血清丙二醛(MDA)、超氧化物歧化酶(SOD)和NO水平,测定线粒体膜电位以及内质网蛋白GRP78蛋白的表达水平,采用TUNEL分析大鼠脑组织细胞凋亡水平,并评价4组大鼠神经功能。结果与模型组比较,低剂量组和高剂量组大鼠SOD和线粒体膜电位水平明显增加,MDA、NO和GRP78蛋白表达水平显著下降,细胞凋亡水平明显下降,神经功能评分显著降低,差异均有显著统计学意义(P<0.05)。低剂量组和高剂量的效果呈剂量依赖性。结论疏血通治疗急性脑梗死缺血再灌注可显著降低氧化应激反应,降低脑组织细胞凋亡。 Objective To explore the effect of Shuxuetong on oxidative stress levels in rats with acute cerebral infarction ischemia-reperfusion. Methods Sixty rats of acute cerebral infarction ischeinia-repeifusion were randomly divided into the model group, low dose group and high dose group. Another twenty rats were selected as sham group. Rats in the low dose group and high dose group were given Shuxuetong 50 mg/kg and 100 mg/kg by intraperitoneal injection. Rats in the model group and control group were given PBS solution by intraperitoneal injection. After treatment of 48 h, MDA, SOD and NO levels in rat serum were determined. Mitochondrial membrane potential and endoplasmic reticulum protein GRP78 were analyzed. The apoptosis of cells was analyzed by TUNEL staining. Neural function in rats of these four groups was analyzed. Results Compared with model group, the SOD and mitochondrial membrane potential levels in rats of the low dose group and high dose group obviously increased, MDA,NO and GRP78 protein expression levels significantly decreased,so did the level of apoptosis and nerve function scores. The difference was of statistical significance (P〈0. 05) . This drug produced effect in a dose-dependent manner in the low dose group and high dose group. Conclusion Shuxuetong can significantly reduce levels of oxidative stress and inhibit brain cell apoptosis in rats with acute cerebral infarction ischemia-reperfusion.
