摘要
为了研究绒山羊皮肤初级毛囊和次级毛囊之间的表达基因模式的差异,筛选影响毛囊发育的重要基因,对6只内蒙古绒山羊阿尔巴斯型个体进行RNA-seq测序,利用生物信息学方法对转录组数据进行分析,在测序结果质量控制后的基础上,对差异表达基因进行筛选、功能注释和富集分析。共筛选到617个在初级毛囊和次级毛囊中差异表达的基因,其中,上调表达基因297个,下调表达基因320个。GO功能注释结果显示,上调表达基因主要行使电子传递链、呼吸链电子传递、线粒体电子传递、基因表达等生物学功能;下调表达基因主要行使细胞吞噬、蛋白质复性、中性粒细胞聚集、细菌真菌防御反应等生物学功能。利用KEGG数据库作为参考,这些基因主要参与半胱氨酸和蛋氨酸代谢、RNA聚合、RNA可变剪切、底物磷酸化等通路。PPP2R2A和CDK2AP1基因可能在皮肤毛囊发育中发挥重要作用,以上结果为今后深入研究皮肤毛囊发育相关基因及功能提供了依据。
This research is conducted to screen differentially expressed genes(DEGs)between the primary and secondary hair follicle(HF)of Cashmere goat's skin to find candidate genes related to development.To analyze the transcriptome data by bioinformatics methods,the RNA-Seq of six individuals from Inner Mongolia Arbas Cashmere goat was performed.On the basis of quality control of the sequencing result,DEGs were identified as well as functional annotation and enrichment analysis was carried out.In this study,617 DEGs has been selected from primary and secondary HF,and among them 297 are up-regulated while 320 are down-regulated.Additionally,the results of gene ontology(GO)expression elucidate that the main function of up-regulated genes are:electron transport chain,respiratory chain electron transport,mitochondrial electron transport,gene expression and other biological functions.However down-regulated expression of genes is mainly exercised as following:cell autophagy,protein complex,neutrophil aggregation,bacterial fungal defense reaction and other biological functions.Using KEGG database as a reference,these genes are mainly involved in the metabolism of cysteine and methionine,RNA aggregation,shear,Parkinson's disease,substrate phosphorylation,and Alzheimer's disease.It will provide a basis for further study on the related genes and functions of hair follicle development in the future.
