竹节参皂苷Ⅳa对高脂饮食小鼠小肠干细胞及其微环境Wnt/β-catenin信号通路的影响 被引量:3

摘要 目的:探讨竹节参皂苷Ⅳa对高脂饮食小鼠小肠干细胞及其微环境Wnt/β-catenin信号通路的影响。方法:C57BL小鼠雌雄各半随机分成4组,每组10只,即正常对照组、模型组及竹节参皂苷Ⅳa低、高剂量组。HE染色观察小肠形态学变化,免疫组化检测各组小肠干细胞数量、PCNA表达及其微环境Wnt/β-catenin信号通路的变化。结果:与正常对照组比较,模型组小肠绒毛长度、V/C值、PCNA表达量及杯状细胞数量、Bmi1阳性表达的小肠干细胞数量显著升高,小肠干细胞微环境Wnt/β-catenin信号显著上调(P<0.05或P<0.01)。与模型组比较,竹节参皂苷Ⅳa组绒毛长度、V/C值、PCNA表达量、杯状细胞数量、Bmi1阳性表达的小肠干细胞数量显著降低,小肠干细胞微环境Wnt/β-catenin信号显著下调(P<0.05或P<0.01)。结论:竹节参皂苷Ⅳa可通过Wnt/β-catenin信号通路调控小肠干细胞微环境,改善其过度增殖分化状态和抑制小肠干细胞增多,调节小肠干细胞功能。 Objective:To investigate the effects of Chikusetsu Saponin Ⅳa(CSIVa) on the small intestinal stem cells(ISCs)and their microenvironment Wnt/β-catenin signaling pathway in mice which fed a high fat diet.Methods:C57BL mice were divided into four groups,with ten mice in each group,the normal control group,model group,the low-dose and high dose CSIVa group.The morphological changes of small intestine was observed by HE staining method.The number of ISCs,and the expression of PCNA,and the change of the microenvironment Wnt/β-catenin signaling pathway was determined by Immunohistochemical staining.Results:Compared with the normal control group,the length of villus length,V/C value,the expression of PCNA,and goblet cell number were increased in the model group,and also increased the number of Bmil positive expression of ISCs,and enhanced the Wnt/β-catenin signaling patway in small intestine (P 〈 0.05 or P 〈 0.01).Compared with the model group,the villus length,the expression of V/C value,PCNA,the number of goblet cells and the number of Bmi1 positive expression ISCs were decreased in CSIVa groups,and down-regulated the Wnt/β-catenin signaling in small intestine (P 〈 0.05 or P 〈 0.01).Conclusion:CSIV a can improve the excessive proliferation and differentiation status of ISCs through the Wnt/β-catenin signaling pathway in the small intestine of mice.
