慢性阻塞性肺疾病患者外周血白细胞线粒体DNA拷贝数变化及其与吸烟的关系

Mitochondria DNA copy number of peripheral blood leucocytes and its relation with smoking in patients with chronic obstructive pulmonary disease

目的探讨慢性阻塞性肺疾病患者外周血白细胞线粒体DNA(mt DNA)拷贝数变化及其与吸烟的关系。方法选择60例COPD患者为COPD组,45例健康人为对照组,提取两组外周血白细胞基因组DNA。比较两组间mt DNA拷贝数,以及COPD组中吸烟与不吸烟患者的mt DNA拷贝数。结果 COPD患者外周血白细胞mt DNA拷贝数低于对照组,吸烟者mt DNA拷贝数低于不吸烟者(均P<0.05)。结论 COPD患者外周血白细胞mt DNA拷贝数的降低,其中吸烟者外周血白细胞mt DNA拷贝数更低。COPD的发生可能与外周血白细胞mt DNA拷贝数减少有关。

Objective To explore the mitochondria DNA（ mt DNA） copy number of peripheral blood leucocytes and its relation with smoking in patients with chronic obstructive pulmonary disease（ COPD）. Methods Sixty patients with COPD were enrolled as COPD group,and 45 healthy individuals as control group. The genomic DNA was extracted from peripheral blood leukocytes in both groups. The mt DNA copy number was compared between the two groups. The mt DNA copy number was also compared between the smokers and the non-smokers of the COPD group. Results The mt DNA copy number of peripheral blood leukocytes in the COPD group was lower than that in the control groups,and the mt DNA copy number in the smokers was lower than that in the non-smokers（ all P 〈 0. 05）. Conclusion The decreased mt DNA copy number of peripheral blood leukocytes can be found in the patients with COPD,and the smoker might have lower mt DNA copy number in peripheral blood leukocytes. The incidence of COPD might correlate with the decrease of mt DNA copy number of peripheral blood leukocytes.
