摘要 目的:建立超高效液相色谱法测定大鼠血浆中多西他赛的质量浓度,为多西他赛的药动学研究提供基础。方法:采用甲醇沉淀法处理大鼠血浆;采用超高效液相色谱法测定多西他赛的质量浓度,色谱柱为Hypersll BDS C18柱(2.1 mm×100 mm,3μm),检测波长为230 nm,流动相为超纯水-甲醇(V∶V=45∶55),流速为0.4 ml/min,柱温为30℃,进样量为10μl。结果:多西他赛和内标紫杉醇的保留时间分别为10.45和7.83 min。多西他赛的线性回归方程为Y=0.101 8 X+0.002 8(R2=0.999 6),其在0.05~10.00μg/ml的范围内线性关系良好,定量下限(S/N=10)为0.05μg/ml。仪器日内和日间精密度均<15%,多西他赛的提取回收率>85%,且在24 h内[-80℃和室温(25℃)条件下]较稳定。结论:本方法简便、快捷、灵敏,可用于大鼠血浆中多西他赛质量浓度的测定。 OBJECTIVE:To establish establish the content of quality of docetaxel in rat plasma by UPLC , so as to provide basis for pharmacokinetic study of docetaxel .METHODS:The blood samples of rats were treated by methanol precipitation , the content of quality of docetaxel was determined by UPLC .The column was Hypersll BDS C 18 (2.1 mm ×100 mm, 3μm) , the detection wavelength was 230 nm, the mobile phase was ultra-pure water-methyl alcohol ( V:V=45:55) with the flow rate of 0.4 ml/min, column temperature of 30 ℃ and sample size of 10 μl.RESULTS:The retention time of docetaxel and internal standard paclitaxel was 10.45 min and 7.83 min, respectively; the standard curve line of docetaxel was Y=0.1018 X+0.0028(R2=0.9996), the linear relationship was good at 0.05-10.00μg/ml;the lower limit of quantification of docetaxel in rat plasma ( S/N=10 ) was 0.05 μg/ml; the intra-day and inter-day precision were less than 15%;the recovery rate was higher than 85%, and the sample was stable at -80℃and room temperature (25 ℃) for 24 hours.CONCLUSIONS:This method is simple, accurate and flexible , which can be used for content determination of quality of docetaxel in rat plasma .
