摘要
基于反向离子电渗透原理,构建无创血糖检测体系。利用丝网印刷工艺制备柔性全固态电极,以普鲁士蓝为电子媒介体,用葡萄糖氧化酶修饰电极制得柔性葡萄糖生物传感器。对传感器的各项修饰条件进行优化,用计时电流法检验其性能,建立起体外葡萄糖溶液定量检测体系。结果表明:在低浓度区对葡萄糖的线性响应范围为0.5~6.5 mmol/L,相关系数(R)为0.993 3,检出限为0.1 mmol/L;高浓度区的线性响应范围为6.5~19.0 mmol/L,相关系数0.992 3。用3D打印制作模拟检测装置,在猪皮上模拟活体电渗透过程进行皮下葡萄糖定量检测。皮下葡萄糖浓度在2.0~14.0 mmol/L范围内,传感器的响应电流与其有较好的相关性(R=0.992 9),浓度范围满足日常血糖检测的需求。为进一步研制实用型无创血糖仪奠定了良好的基础。
Based on reverse iontophoresis principle,a non-invasive blood glucose detection system was built up.Flexible screen printed solid electrodes,using Prussian blue as the medium,combined with glucose oxidase,formed the flexible glucose biosensor.The performance of the biosensor was evaluated by chronoamperometry.The glucose in vitro quantitative detection system was established by optimizing each of modified conditions.Within the scope of low concentration,the linear range of glucose concentration was 0.5-6.5 mmol/L,the correlation coefficient(R) was 0.993 3 and detection limit was 0.1 mmol/L,and within the scope of relatively high concentration,the linear range was 6.5-19.0 mmol/L and the correlation coefficient was 0.992 3.A 3D printed device was designed to simulate reverse iontophoresis on pig skin,thus the quantitative detection system of subcutaneous glucose was established.The responseof the sensor had a good correlation with the subcutaneous glucose concentration in the range of 2.0-14.0 mmol/L(R=0.992 9) of daily blood glucose testing.The results found the basis for the development of a practical non-invasive blood glucose meter.
