复方湿生扁蕾提取液镇痛、抗炎及急性毒性研究 被引量:9

摘要 目的研究复方湿生扁蕾提取液的镇痛、抗炎作用,并考查其单次给药的急性毒性反应,为临床用药提供一定的理论依据。方法通过热板致小鼠疼痛、二甲苯致小鼠耳肿胀模型,观察复方湿生扁蕾提取液对小鼠的镇痛、抗炎作用;通过单次以最大灌胃体积不同浓度给药观察该复方提取液对小鼠的急性毒性。结果本复方提取液具有一定的镇痛、抗炎作用,复方组给药前后的痛阈差值与空白对照组相比具有显著性差异(P<0.01),复方组对二甲苯致小鼠耳肿胀抑制率达31.7%。100、120 g·kg-1剂量组给药后4 h之内部分小鼠出现昏睡、萎靡少动、进食减少、稀便、反应迟钝等临床症状,这些症状在2 d内逐渐消失。观察期间这两复方组体重增长缓慢,观察结束后剖检主要脏器未见肉眼可见病变。结论在100、120 g·kg-1剂量下发现对小鼠体重具有不良影响,未发现致小鼠死亡及器官性病变的毒性作用,其LD50大于120 g·kg-1,安全性较好,可一定程度上为临床用药提供实验依据。 Objective To study the anti-inflammatory and analgesic effects of the extraction of compound Gentianopsis paludosa,and examine the acute toxicity of single drug reaction,to provide a theoretical basis for clinical medication.Methods The mice pain caused by hot plate,ear swelling caused by xylene were applied to observe the anti-inflammatory and analgesic effects of compound Gentianopsis paludosa extract on mice.By one-time intragastric administration of mice to observe the acute toxicity of the compound extract liquid on mice. Results Certain analgesic and anti-inflammatory effects were demonstrated in the extract of this compound,and analgesic effects of the compound group and control group difference threshold before and after treatment compared with significant difference( P<0.01),the inhibition rate of xylene induced ear swelling in compound group was 31.7%.Within 4 h after 100 and 120 g·kg-1 dose,some mice showed lethargy,malaise,less feeding,watery stools,unresponsive and other clinical symptoms,these symptoms gradually disappeared within 2 d.During the observation period,the weight of the two compound group was increased slowly.After the observation,the main organs in the necropsy showed no visible pathological changes.Conclusion The adverse effects in the weight of mice was observed at 100,120 g·kg-1 dose,no toxic effect caused by the death of mice and organ lesions.The LD50 value was greater than 120 g·kg-1,and the safety was better,It can provide experimental basis for clinical medication to a certain extent.
