摘要
采集临床疑似伪狂犬病例病料并分离到1株病毒。PCR鉴定仅能扩增出伪狂犬病毒特异性条带,其他常见的7种猪群疫病病毒为阴性。家兔接种试验能引起典型的神经症状,最终衰竭死亡,证实所分离病毒为强毒株,将分离到的伪狂犬病毒命名为HX14株。选取我国不同时期分离的伪狂犬野毒gE基因进行序列比对分析,结果显示,HX14株与参比序列同源性在93.8%~97.2%之间,同源性最高的是Min-A株,最低的是GZ-Z1株。氨基酸序列比较,HX14株和Ea、GDSH、HB-HS、HB-LF、HBJZ、LXB6、LXB88、Min-A、P-PrV、Yangsan株的同源性均为93.2%,与89V87同源性最低,为88.1%。HX14株在8个氨基酸位点上和参考毒株明显不同,分别是P^(12)→M^(12)、^(14)LRREPP19→^(14)CSARAA19、T^(115)→R^(115)。研究表明HX14株为gE基因变异较为明显的伪狂犬野毒强毒株。
A stain of pseudorabies virus was isolated from tissues of porcines suspected with Aujeszky's disease.Specific bands of pseudorabies virus were found by PCR,yet no bands were found using specific primers for seven other pig viruses.Rabbits appeared neurological symptoms even to death after being challenged with the virus suspension.It confirmed that the isolated virus was a virulent strain of pseudorabies virus,and it was named HX14 strain.gE gene sequencing results showed that the homologies between HX14 and wild pseudorabies viruses Min-A,Ea,GDSH,HB-HS,HB-LF,HBJZ,LXB6,LXB88,PPrV,Yangsan were 93.8%-97.2%.The highest homology was Min-A strain and the lowest was GZ-Z1.The result of amino acid sequence comparison with Ea,GDSH,HB-HS,HB-LF,HBJZ,LXB6,LXB88,MinA,P-PrV and Yangsan showed that the homology were 93.2%.The lowest homolog was 88.1%compared with the 89V87.It was significantly different on eight amino acid sites,P^(12)→ M^(12),^(14) LRREPP19→14CSARAA19and T^(115)→R^(115),between HX14 and the reference strain.Taken together,the study showed that HX14 was a virulent strain of pseudorabies virus with obvious mutation in gE gene.
