三氯苯达唑、吡喹酮治疗大鼠感染斯氏狸殖吸虫的实验研究

Experimental study of triclabendazole and praziquantel on the treatment of rats with Pagumogonimus skrjabini

摘要 目的观察三氯苯达唑、吡喹酮杀灭斯氏狸殖吸虫的疗效及对虫体体壁超微的结构改变。方法 15只大鼠随机分成三氯苯达唑组、吡喹酮组和对照组。经腹腔注射斯氏狸殖吸虫囊蚴感染大鼠。感染60 d后两治疗组治疗、对照组不作治疗。用药3 d后剖杀大鼠,计数虫体回收数,计算检虫率及减虫率,并与对照组比较;另外,检获虫体作透射电镜观察。结果三氯苯达唑组和吡喹酮组检虫率分别为(6.69±5.95)%和(22.60±8.95)%,与对照组(53.33±9.43)%比较,差异有统计学意义(P=0.000);三氯苯达唑组和吡喹酮组减虫率分别为87.50%和57.50%,差异有统计学意义(P=0.003)。透射电镜观察:三氯苯达唑组虫体皮层消失,基质膜破坏,肌层坏死;吡喹酮组虫体皮层受损,肌层紊乱;对照组体壁结构完整。结论三氯苯达唑杀灭斯氏狸殖吸虫效果优于吡喹酮,而且三氯苯达唑对虫体体壁造成损伤程度比吡喹酮严重。 Objective To observe the therapeutic effects between triclabendazole and praziquantel on rats with Pagumogonimus skrjabini and the ultrastructural changes on Pagumogonimus skrjabini caused by them. Methods 15 rats were randomly divided into triclabendazole group, praziquantel group and control group. They were each injected through abdominal cavity with metacercariaes of P skrjabini. Two treatment groups were treated on the 60th days after infection. Rats were dissected on the 3rd day after treatment. Counting No of worms recovered, calculating rate of worms recovered and reduction rate of worms, and comparing with rats of control group, and the worms recovered were observed under transmission electron microscope. Results The rate of worms recovered among triclabendazole group, praziquantel group and control group were (6. 69 ± 5.95 )%, (22. 60 ±8. 95)% and (53. 33 ±9. 43)% respectively, there were significant among them (P = 0. 000) ; the reduction rate of worms between triclabendazole group and praziquantel group were 87. 50% and 57. 50% respectively, there were significant between them( P = 0. 003 ). The observation of transmission electron microscope : cortexes of worm of triclabendazole group were disappear and base material membranes were broken, and muscle layers were perished, cortexes of worm of praziquantel group were damaged, and the muscle layer is not orderly. Body walls structures of worms in control group were completely. Conclusion The therapeutic effects of triclabendazole were better than that of praziquantel, and the damage degree of body walls of P skrjabini caused by triclabendazole was worse than that of praziquantel.
