摘要 目的比较两种不同兔血瘀模型的血液流变学指标及凝血参数的差异性,探讨各个模型的特点及适用性。方法分别以右旋糖酐和肾上腺素制备两种兔血瘀模型,比较不同模型全血黏度(WBV)、血浆粘度、红细胞聚集指数、血红蛋白(HB)、红细胞压积(HCT)、纤维蛋白原(FIB)含量、凝血酶原时间(PT)、活化部分凝血酶时间(APTT)和凝血酶时间(TT)的差异性,对两种模型的血瘀指标进行评价。结果与给药前相比,肾上腺素血瘀模型能显著升高全血黏度在1/s、5/s、30/s、150/s、200/s的切变率、凝血酶原时间、部分凝血活酶时间以及红细胞聚集指数(P<0.01,P<0.05),差异均有统计学意义,此模型适用于各类活血化瘀药物的研究;右旋糖酐血瘀模型能显著升高全血粘度在1/s、5/s、30/s的切变率、纤维蛋白原以及红细胞聚集指数(P<0.01,P<0.05),但部分凝血活酶时间和凝血酶时间显著低于给药前,差异有统计学意义(P<0.01),此模型对微循环影响颇大,适合应用于改善微循环药物的研究。结论不同造模方法建立的兔血瘀模型在血液流变学指标及凝血参数上的影响不同,为研究不同活血化瘀药物精确选择血瘀证模型提供了参考。 Objective To compare the hemorheology indexes and coagulation parameters of two rabbit modelsofblood stasis syndrome which were established by using two different methods, and to discuss thecharacteristics and applicability of each rat model. Methods Two different rabbit models of blood stasissyndrome were established including adrenaline group and dextranum group. The differences in whole blood viscosity (WBV), plasma viscosity, erythrocyte aggregation index, hemoglobin (HB), hematocrit(HCT), fibrinogen(FIB) content, prothrombin time(PT), activated partial thromboplastin time(APTT) and thrombin time(TT)were compared among all groups. Results The hemorheology indexes were more significant in adrenaline group with higher applicability, especially the shear rate of whole blood viscosity in 1/s, 5/s, 30/s, 150/s, 200/s(P 〈0.01, P 〈 0. 05 ). The model used in the research of all kinds of promoting blood circulation to remove blood stasis drugs. Dextran model of blood stasis can significantly affect fibrinogen, activated partial thromboplastin time and thrombin time, erythrocyte aggregation index, whole blood viscosity shear rate (P 〈 0. 01, P 〈 0. 05). But the partial thromboplastin time and thrombin time were significantly lower than that before treatment, the difference was statistically significant (P 〈 0. 01 )The model can be applied to improve microcirculation in drug research. Conclusion The rabbit models of blood stasissyndrome established by using different methods show different characteristics in hemorheology indexes and coagulation parameters, and have different applicability. This study provides some reference for choosing rabbit models of blood stasis syndrome in the research on the activities of medicinals with the actions of activating blood and resolving stasis.
