摘要 目的检测基质金属蛋白酶-11(MMP-11)和转录因子Sp1在结肠癌中的表达,探讨二者表达的相关性及临床意义。方法采用免疫组织化学方法检测72例结肠癌患者肿瘤组织及20例癌旁正常组织中MMP-11和转录因子Sp1的表达。结果 MMP-11和转录因子Sp1在72例结肠癌中表达的阳性率分别为63.89%(46/72)和66.67%(48/72),与在20例正常结肠组织中表达阳性率(分别为20%和10%)的差异有统计学意义(P<0.05)。MMP-11的过表达与结肠癌的TNM分期、分化程度及淋巴结转移相关(P<0.05),而与患者的年龄、性别、肿瘤大小无明显相关性;在结肠癌组织中MMP-11和转录因子Sp1的表达之间呈明显正相关(r=0.45,P<0.01)。结论 MMP-11和转录因子Sp1与结肠癌的生物学行为密切相关,可作为预测结肠癌浸润和转移潜能及评估预后的参考指标之一。 Objective To determine the expressions of matrix metalloproteinase-11(MMP-11) and transcription factor Sp1 in colorectal carcinoma,and to investigate their relations. Methods Immunohistochemical technique was used to determine the expressions of MMP-11 and transcription factor Sp1 in 72 specimens of colorectal cancer tissues and 20 specimens of adjacent normal colorectal tissues. Results The positive expression rates of MMP- 11 and transcription factor Sp1 in colorectal cancer tissues were 63.89%(46/72) and 66.67%(48/72),and those in adjacent normal colorectal tissues were 20% and 10%(P〈0.05). The expressions of MMP-11 in colorectal cancer tissues were correlated with TNM stage,differentiation degree and lymph node metastasis(P〈0.05),but there was no correlation with age,sex and tumor size. MMP-11 expression was positively correlated with transcription factor Sp1 expression in colorectal cancer tissues(r=0.45,P〈0.01). Conclusions MMP-11 and transcription factor Sp1 have close relations with the biological behavior of colorectal carcinoma,and it may be markers in predicting the invasion,metastasis and prognosis of colorectal carcinoma.
