摘要
目的:通过建立实验性精索静脉曲张大鼠动物模型,探讨精索静脉高位结扎术对同侧睾丸生精细胞凋亡机制的影响。方法:雄性大鼠60只,随机分为对照组、假手术组、EV组(模型组)、Varicocelectomy组(治疗组),每组15只。术后取左侧睾丸,采用HE染色观察睾丸组织学变化,TUNEL法检测生精细胞凋亡指数,免疫组化和RT-PCR法分别检测Bcl-2、Bax和Caspase-3的表达。结果:模型组大鼠左侧睾丸组织可见生精上皮排列紊乱,生精细胞广泛脱落,细胞凋亡指数明显高于对照组、假手术组(P<0.05),治疗后凋亡指数明显减少,与模型组比较,差异有显著性(P<0.05);治疗组中Bax、Caspase-3表达减少,Bcl-2表达增加,与模型组比较,差异有显著性(P<0.05)。结论:精索静脉高位结扎术可抑制睾丸生精细胞凋亡,且Bcl-2、Bax和Caspase-3通过线粒体信号转导通路参与并调控生精细胞凋亡过程,从而在精索静脉曲张疾病中发挥保护作用。
Objective: To investigate the effect of varicocelectomy on the mechanism of spermatogenic cell apoptosis in ipsilateral testis through the establishment of experimental varicocele model in rats. Methods: Sixty male rats were randomly divided into four groups with 15 animals each.con- trol group, sham-operation group, EV group (varicocele model group), and varicocelectomy group (treatment group). HE staining was used to observe the histological changes of left testis, and the TUNEL assay was used to measure the apoptotic index of spermatogenic cells. The ex- pression of Bcl-2, Bax and Caspase-3 were detected by immunohistochemistry and RT-PCR meth- od. Results: The spermatogenic cells were arranged in disorder and widely shed. In the EV group, the apoptotic index was significantly higher than those in the control group and sham-op- eration group (P〈0.05). After treatment, compared with EV group, the apoptotic index was significantly reduced, and there were statistically differences between the two groups (P〈0.05) ; Compared with EV group, the expression of Bax and Caspase 3 decreased, and Bcl-2 increased inthe varicocelectomy group,and the differences were significant lectomy inhibits the spermatogenie cell apoptosis that may Caspase-3 through mitochondrial signal transduction pathway, process of varicoeele.
