小剂量IL-2对原发性干燥综合征患者外周血CD4^+T细胞的影响

Low-dose of interleukin-2 could affect levels of cytokines secreted by CD4^+T cells in the peripheral blood of primary Sj?gren's Syndrome patients

目的了解小剂量IL-2对原发性干燥综合征(primary Sj?gren's Syndrome,pSS)患者外周血中CD4^+T细胞分泌IL-6、IL-8和IL-10等细胞因子的影响。方法选取2016年6月至2016年12月就诊于本科的21例p SS患者及20例健康对照人员。化学发光法检测患者和对照组血清中IL-6、IL-8和IL-10的水平。磁珠分选患者外周血CD4^+T细胞,分别以不同浓度的IL-2进行刺激,培养72小时后检测细胞上清液中IL-6、IL-8和IL-10的水平。结果 pSS患者血清中IL-6和IL-8水平显著高于对照组(P值分别为P<0.01,P<0.05),IL-10水平与对照组无明显差异。经过小剂量IL-2刺激后,CD4^+T细胞上清液中的炎性因子IL-6和IL-8较前明显下降(P值分别为P<0.05,P<0.001),而抗炎因子IL-10有明显上升(P<0.05)。结论小剂量IL-2能够明显影响干燥综合征患者外周血中CD4^+T细胞分泌细胞因子的水平,对免疫系统可能有负调节作用。

Objective To investigate the effect of low-dose IL-2 on cytokines secretion by CD4＋T cells of patients with primary SjSgren＇s Syndrome （pSS）. Methods Twenty-one pSS patients from June to December 2016 as well as 20 healthy persons were enrolled. Levels of IL-6,IL-8 and IL-10 in sera were measured. CD4＋T cells selected by MACS from peripheral blood was cultured and stimulated with different concentrations of low-dose IL-2 for 72 hours. Levels of IL-6,IL-8 and IL-10 in the supernatant were detected. Results Levels of IL-6 and IL-8 in sera from pSS were significantly higher than those in control group （P〈0.01and P〈0.05, respectively）. After stimulation with IL-2, levels of IL-6 and IL-8 in the supernatant were significantly decreased （P〈0.05 and P〈0.001, respectively）, while the level of IL-10 got increased （P〈0.05）. Conclusion Low-dose of IL-2 could significantly affect cytokines secretion by CD 4＋T cells in pSS patients, and may play a role in the negative regulation of the immune system.
