摘要 目的:探究清热败毒合剂对难愈性创面金黄色葡萄球菌、铜绿假单胞菌细菌生物膜的作用。方法:将50只SD大鼠随机分为空白对照组、清热败毒合剂组、五味清毒饮组、阿莫西林组和头孢哌酮组,每组10只,分别予生理盐水、清热败毒合剂、五味消毒饮、阿莫西林和头孢哌酮灌胃,连续3 d,制备药物血清。分光光度法检测药物血清对于金黄色葡萄球菌和铜绿假单胞菌细菌增殖的影响;结晶紫染色法和CONA-FITC/PI法检测药物血清对细菌生物膜形成的影响;生物发光法检测药物血清对于细菌自诱导分子AI-2的影响。结果:清热败毒合剂能够通过降低难愈性创面金黄色葡萄球菌和铜绿假单胞菌AI-2浓度,抑制细菌生物膜的形成,从而抑制细菌的增殖,促进创面愈合。结论:清热败毒合剂对慢性难愈性创面金黄色葡萄球菌、铜绿假单胞菌细菌生物膜的形成具有抑制作用。 Objective: To study the effect of Qingre Baidu Mixture on the biofihn of Staphylococcus aureus and Pseudomonas aeruginosa in chronic and refractory wounds. Methods: A total of 50 SD rats were randomly divided into blank control group, Qingre Baidu mixture group, Wuwei Xiaodu Drink group, amoxicillin group and Cefoperazone group, 10 rats in each group, and the rats were administrated with normal saline, Qiugre Baidn mixture, Wuwei Xiaodu Drink, amoxicillin and Cefoperazone respectively, for three days, to prepare drug serum. The efi^ets of drug serum on the proliferation of Staphylococeus aureus and Pseudmnonas aeruginosa were de- tected by spectrophotometry; The effect of drug serum on formation of bacteria biofilm were evaluated by crystal violet and CONA-FITC/PI staining, and the effect on AI-2 secreted by bacteria were detected by biolumines- cence assay. Results: Qingre Baidu mixture couht reduce the Al-2 concentration from Staphylococcus aureus and Pseudomonas aeruginosa in chronic and refractory wounds, inhibit the formation of bacterial biofilm, thereby in- hibit the proliferation of bacteria, and promote wound healing. Conclusion: Qiugre Baidu mixture could inhibit the formation of biofihn of Staphylococcus aureus and Pseudomonas aeruginosa in chronic and refractory wonnds.
