摘要
目的:观察愈癫汤对MK-801致谷氨酸功能低下精神分裂症模型大鼠认知与学习记忆功能的影响。方法:SD雄性成年大鼠48只,随机分为空白组、模型组、利培酮组及愈癫汤组。其中模型组、利培酮组及愈癫汤组应用MK-801腹腔注射造模,空白组应用0.9%Na Cl溶液行相同方法腹腔注射,连续14 d。造模结束,于第15天起,利培酮组给予0.036 g/L利培酮悬浊液灌胃,愈癫汤组给予浓度0.92 g/ml愈癫汤水煎剂灌胃,空白组、模型组给予相应体积蒸馏水灌胃,连续14 d。应用Morris水迷宫进行行为学评价,观察并比较各组受试大鼠认知及学习记忆能力。结果:学习第4天、第5天,模型组大鼠逃避潜伏期较空白组明显延长(P<0.05,P<0.01);与模型组比较,愈癫汤组大鼠逃避潜伏期时间明显缩短(P<0.05)。愈癫汤组大鼠各时间点逃避潜伏期呈下降趋势,在学习第2天起即呈现显著下降趋势(P<0.05,P<0.01)。模型组大鼠穿越安全平台次数、中央活动路程比及中央活动时间比较空白组明显降低(P<0.01);与模型组比较,愈癫汤组大鼠穿越安全平台的次数明显增加,中央活动路程比及中央活动时间比均明显升高(P<0.05,P<0.01)。结论:愈癫汤能够改善精神分裂症模型大鼠认知及学习记忆功能,并缓解其焦虑、恐惧症状。
Objective: To observe the effects of Yudian Decoction on cognition and learning memory function in model rats of schizophrenia with glutamate hypofunction induced by MK-801. Methods: 48 male adult SD rats were randomly divided into the blank group, model group,risperidone group and Yudian Decoction group. The rats in the blank group were treated with 0.9% NaCl solution by intraperitoneal injection, and the rats in the other groups were treated with MK-801 by intraperitoneal injection. The modeling course was continuous 14 days, and the experimental animal model of schizophrenia with glutamate hypofunction was established. After modeling, from the fifteenth day, the risperidone group was treated with risperidone suspension at concentration of 0. 036 g/L, the Yudian Decoction group was treated with Yudian Decoction at concentration of 0.92 g/ml,and the blank and model group were treated with distilled water at the corresponding volume. After intragastric administration for continuous 14 days, the behavioral evaluation was performed by Morris water maze, and the cognition and learning memory abilities of rats in each group were observed and compared. Results : On the fourth and fifth day of learning experiment,compared with the blank group,the escape latency time of rats was obviously prolonged in the model group(P 〈 0.05 ,P 〈 0.01 );compared with the model group, the escape latency time of rats was obviously shortened in the Yudian Decoction group(P 〈0.05). The escape latency time of rats in the Yudian Decoction group showed the decreasing trend at each timepeint, and there was the significant decreasing trend from the second day of learning( P 〈 0.05, P 〈 0.01 ). Compared with the blank group, the platform crossing frequency, the distance ratio of central activity and time ratio of central activity were obviously decreased in the model group(P 〈 0.01 );compared with the model group,the platform crossing frequency,the distance ratio of central activity and time .ratio of central activity were obviously increasd , in the Yudian Decoction group(P 〈 0.05 ,P 〈0.01 ). Conclusion: Yudian Decoction can improve the cognition and learning memory function in model rats of schizophrenia, and eliminate the anxiety and fear state.
