摘要
目的:探讨胆仙咳喘宁胶囊治疗慢性支气管炎(慢支)的作用机制。方法:48只SD大鼠随机分为空白对照组、模型组、阳性对照(DXM)组及胆仙咳喘宁高、中、低剂量组。除空白对照组外,均采用改良烟熏法复制慢支模型。各组大鼠分别用DXM、胆仙咳喘宁及0.9%Na Cl溶液灌胃干预,连续干预21 d。采用免疫组化染色方法(PV二步法)测定各组大鼠肺组织中NF-κB(p65)表达;采用固相夹心法酶联免疫吸附实验(ELISA)测定各组大鼠的血清及肺组织中的肿瘤坏死因子-α(TNF-α)含量。结果:模型组大鼠肺组织中NF-κB(p65)表达较空白对照组明显升高(P<0.01);胆仙咳喘宁高、中、低剂量组及DXM组大鼠肺组织中NF-κB的表达显著低于模型组(P<0.01,P<0.05)。模型组大鼠血清和肺组织中TNF-α水平较空白对照组均明显增加,胆仙咳喘宁高、中、低剂量组及DXM组大鼠血清及肺组织中TNF-α含量均明显低于模型组(P<0.05,P<0.01)。结论:胆仙咳喘宁胶囊可能通过抑制NF-κB的活化抑制炎症因子TNF-α的表达。
Objective: To investigate the mechanism of Danxian Kechuanning Capsule in the treatment of chronic bronchitis(CB). Methods: 48 SD rats were randomly divided into the blank control group, model group, dexamethasone (DXM) group and Danxian Kechuanning Capsule(DXKCN) groups with high-, middle- and low-dose. Except the blank control group, the modified method of smoking inhalation was used to replicate the CB model. The rats in each group were treated with the corresponding drugs or 0.9% NaCl solution by intragastric administration for continuous 21 days. The expression of NF-KB p65 in lung tissue of rats was detected by immunohistochemical staining ( PV two-step method ), and the content of tumor necrosis factor-α (TNF-α) in serum and lung tissue of rats was determined by solid phase sandwich enzyme-linked immunosorbent assay (ELISA). Results: Compared with the blank control group, the expression of NF-κB p65 in lung tissue of rats was obviously increased in the model group ( P 〈 0. 01 ) ;compared with the model group, the expression of NF-κB in lung tissue of rats was significantly decreased in the DXKCN groups with high-, middle- and low-dose and DXM group ( P 〈 0.01, P 〈 0.05 ). Compared with the blank control group, the level of TNF-α in serum and lung tissue of rats was obviously increased in the model group ; compared with the model group, the level of TNF-α in serum and lung tissue of rats was obviously decreased in the DXKCN groups with high-, middle- and low-dose and DXM group ( P 〈 0.01, P 〈 0. 05 ). Conclusion: Danxian Kechuanning Capsule may inhibit the expression of inflammatory faetor TNF-ot by suppressing the activation of NF-κB.
