重组人促红细胞生成素对心梗大鼠HIF-1α与Survivin表达的影响 被引量:1

The effect of recombinant human erythropoietin on the expressions of HIF-1α and Survivin in myocardial infarction rats

摘要 目的探讨重组人促红细胞生成素(rHuEPO)对心梗大鼠心肌缺氧诱导因子-1α(HIF-1α)与生存素(Survivin)表达的影响。方法 45只SD大鼠,随机分为假手术组、心肌梗死组和rHuEPO治疗组。用Western blot方法检测各组大鼠梗死区周围心肌中HIF-1α与Survivin的表达水平,RT-PCR方法检测梗死区周围心肌HIF-1αmRNA与Survivin mRNA表达变化。结果心肌梗死组大鼠心肌梗死区周围心肌HIF-1α蛋白与mRNA的表达水平显著高于假手术组,而Survivin蛋白与mRNA的表达水平则显著降低(P<0.01);与心肌梗死组相比,rHuEPO治疗组心肌梗死区周围心肌HIF-1α蛋白与mRNA的表达水平明显降低,而Survivin蛋白与mRNA的表达水平则显著升高(P<0.01)。结论 rHuEPO能够抑制心梗大鼠心肌梗死区周围心肌HIF-1α过表达并上调Survivin的表达。 Objective To study the effect of recombinant human erythropoietin(rHuEPO) the expressions of HIF-1 α (Hypoxia-inducible factor-1 α ) and Survivin in myocardial infarction rats. Methods 45 SD rats were randomly divided into the sham group, the myocardial infarction gronp(MI group) and recombinant human erythropoietin group(rHuEPO group). The expressions of HIF-1 eL and Survivin proteins and mRNAs in parafarction area were examined by Western blot and RT-PCR respectively. Results The expression levels of HIF-1α protein and mRNA in parafaretion area were increased significantly in MI group than in sham group, but decreased in rHuEPO group than in MI group(P 〈 0.01), The expression levels of Survivin protein and mRNA in parafarction area were decreased significantly in MI group than in sham group, but increased in rHuEPO group than in MI group(P 〈 0.01). Conclusion rHuEPO could down-regulate the expression of HIF-1α protein and mRNA and up-regulate the expression of Survivin protein and mRNA in parafarction area of myocardial infarction rats.
