三种温度保存实验室常见菌种的比较 被引量:3

Comparison of laboratory common bacteria in three kinds of temperature preservation

摘要 目的探索一套简便且行之有效的实验室菌种保存方法。方法采用三种常见实验室保存菌种的温度作比较。收集从食品中分离的菌种26株,参与国家级质控考核及能力验证收集菌种12株,从国家菌种保藏中心购买12株,均为非苛养菌。观察采用4℃、-20℃、-70℃三种温度分别保存1个月、6个月、12个月、24个月、36个月的存活率。结果保存1个月后菌种的存活率分别为98.0%、100.0%、100.0%,保存6个月后菌种的存活率分别为90.0%、100.0%、100.0%,保存12个月后菌种的存活率分别为76.0%、100.0%、100.0%,保存24个月后菌种的存活率分别为56.0%、90.0%、98.0%,保存36个月后菌种的存活率分别为38.0%、64.0%、94.0%。结论 4℃、-20℃、-70℃在一定时间内均能很好的保存实验室的常见菌,其中-20℃和-70℃在长期保存菌种时明显优于4℃。 Objective To find a simple and effective method for preservation of laboratory bacteria.Methods The temperatures of three common laboratory preserved bacteria strains were compared. 26 strains isolated from food,12 strains of bacteria participated in the national quality control assessment and capacity testing were collected,12 strains of bacteria were bought from the national bacteria collection center,all of them were non harsh bacteria. The survival rates of 1 month,6 months,12 months,24 months and 36 months were observed when strains were respectively stored at 4 ℃,-20 ℃,-70 ℃.Results After 1 month of preservation,the survival rates of the strains were 98.0%,100.0%,100.0%respectively. After 6 months of preservation,those were 90.0%,100.0%,100.0%. After 12 months of preservation,those were 76.0%,100.0%,100.0%. After 24 months of preservation,those were 56.0%,90.0%,98.0%,and after 36 months of preservation,those were 38.0%,64.0%,94.0%. Conclusion The common bacteria in laboratory can be well preserved at 4 ℃,-20 ℃ and-70 ℃ for a certain time.-20 ℃and-70 ℃ are obviously better than 4 ℃ in long-term preservation of strains.
