龙灯胶囊对大鼠心肌缺血再灌注损伤的保护作用

Protective Effect of Longdeng Capsule on Myocardial Ischemia Reperfusion Injury in Rats

目的:观察龙灯胶囊(LDJN)对大鼠心肌缺血再灌注损伤的保护作用。方法:将80只SD大鼠,随机分为正常组,模型组,灯盏花素提取物、地龙提取物和LDJN三种药物的低(170mg/kg·d)、高(680mg/kg·d)剂量组,每组10只,其中灯盏花素提取物为阳性对照组。结扎各组大鼠冠状动脉左前降支(LAD)40min,松扎再灌注120min,以复制心肌缺血再灌注损伤模型。造模成功后,采用红四氮唑(TTC)染色测量大鼠心肌梗死面积,以图像处理软件(IPP)测定大鼠心肌缺血面积的改变。结果:与模型组比较,灯盏花素提取物(170、680mg/kg)和地龙提取物(170、680mg/kg)的缺血面积百分比明显缩小(P<0.01);地龙提取物(170mg/kg)的心肌梗死面积明显缩小(P<0.01),LDJN(680mg/kg)与模型组比较差异无统计学意义(P>0.05)。结论:LDJN可以通过减小心肌梗死面积,发挥对大鼠心肌缺血再灌注损伤的护作用。

ObjectiveTo observe the protective effect of Longdeng capsule （LDJN） on myocardial ischemia reperfusion injury in rats. Methods Totally 80 SD rats were randomly divided into control group, model group, Dengzhanhuasu extract 170, 680mg/kg control groups, earthworm extract 170, 680mg/kg treated groups, and LDJN 170, 680mg/kg treated groups （n = 10） . The myocar- dial ischemia reperfusion injury model was established through occluding the left anterior descending branch of coronary artery for 40 min and removing the ligation later to reperfuse for 30 min in vivo. Different doses of aqueous extract were intragastrieally administered once per day for 4 days before operation. Using TTC （TFC） staining measure myocardial infarct size, and image processing software （IPP） was used to gauge the size vary from myocardial ischemia area. Results Compared with myocardial isehemia reperfusion control group, the percentage of ischemic area with Dengzhanhuasu extract 170, 680mg/kg groups and earthworm extract 170, 680mg/kg groups were significantly decreased （P 〈 0.01 ） ; myocardial infarct size of LDJN 170mg/kg was significantly decreased （P 〈 0. 01 ） . Conclusion LDJN has protective effect on myocardial ischemia reperfusion injury in rats by reducing the myocardial infarct area.
