Box-Behnken效应面法优化泰山紫草中总萘醌提取工艺

摘要 目的优选泰山紫草总萘醌的提取工艺。方法在单因素试验基础上,采用Box-Behnken效应面法,以总萘醌的提取率为指标,考察乙醇体积分数、提取时间、料液比等的影响,利用紫外分光光度计测定总萘醌的含量。结果确定泰山紫草中总萘醌的最佳工艺为:乙醇体积分数80%,提取时间30 min,料液比1∶18,提取2次,总萘醌的提取率为3.384%。结论 Box-Behnken效应面法用于泰山紫草中总萘醌提取工艺条件的优选是可行的,模型预测效果较好。 Objective: To optimize the extraction technology for total naphthoquinones from Lithospermum erythrorhizon Sieb. et Zucc by Box-Behnken design. Methods: Based on the results of single-factor experiment,Box-Behnken experimental design combined with response surface methodology was employed,investigatine the ethanol concentration,solid-liquid ratio,and extraction time affecting the extraction process,the extraction rate of total naphthoquinones indicators. The total naphthoquinone content by UV spectrophotometer. Results: The optimization processing parameters was as follows: ultrasonic extracted 30 min with 18 times the amount of 80% ethanol and extract twice,respectively. The extraction rate of total naphthoquinones was 3. 384%. Conclusion: Box-Behnken experimental designis feasible,and highly predictive.
