摘要
目的探讨桔梗总皂苷对2型糖尿病大鼠的治疗作用。方法 100只雄性SD大鼠随机分为正常对照组(NCG),模型对照组(MCG),以及桔梗总皂苷低、中和高剂量治疗组(PLG,PMG和PHG),每组20只。造模成功后,PLG,PMG和PHG组给予桔梗总皂苷100,200和400mg·kg^(-1)·d^(-1)灌胃,NCG和MCG组给生理盐水灌胃,治疗4周和8周后处死大鼠各半,免疫组和Western Blot检测肝脏SREBP1c和ACCα的表达。结果 MCG组较NCG组TG和FFA明显升高(P<0.05),治疗4周后明显降低(P<0.05),治疗8周后进一步降低;免疫组化和Western Blot可见MCG组SREBP1c和ACCα表达明显升高(P<0.05),治疗4周后没有改善,治疗8周后明显降低(P<0.05)。结论桔梗总皂苷对2型糖尿病大鼠TG和FFA有明显的改善作用,这可能与其可以降低肝脏中SREBP1c和ACCα的表达有关。
Objective To study platycodins on the effection in liver of on type 2 diabetic rat.Methods 100 male SD rats were randomly divided into normal group(NCG), model group(MCG), and platycodins low,medium and high dose group(PLG, PMG and PHG),(n =20). PLG, PMG and PHG group were given of platycodins 100,200 and 400 mg·kg^(-1)·d^(-1) by gavage,NCG and MCG group received saline or all.After 4 weeks and 8 weeks, rats were sacrificed, comparated the results of immunohistochemistry and Western Blot.Results MCG group than in NCG group,TG and FFA were significantly higher(P<0.05).After 8 weeks of treatment,it was significantly lower(P<0.05), immunohistochemistry and Western Blot could been that found MCG group SREBP1 c and ACCα was significantly lower(P<0.05),After 8 weeks of treatment,it were significantly improved(P<0.05).Conclusion Platycodin to TG and FFA have significant improvement,which may be related to improve the expression of SREBP1 c and ACCα.
