mir-638和Sox2在原发性肝癌中的表达及临床意义

Expression and clinical significance of Mir-638 and Sox2 in in hepatocellular carcinoma

目的检测mi R-638和Sox2在原发性肝细胞癌中的表达情况,探讨其与肝癌患者临床病理特征的关系。方法采用实时定量PCR(q RT-PCR)对78例原发性肝癌患者癌组织及对应癌旁组织中mi R-638和Sox2 m RNA表达水平进行检测。免疫组化法检测所有癌组织及其相应的癌旁正常组织Sox2蛋白的表达情况。分析mir-638和Sox2与肝癌患者临床病理参数间的关系。结果 (1)与癌旁正常组织相比,mi R-638在肝癌组织中的表达明显降低(P<0.05),而Sox2 m RNA在癌组织中的表达显著升高(P<0.05);(2)Spearman相关分析显示,在肝癌组织中mi R-638与Sox2蛋白的表达呈显著负相关(r=-0.441,P<0.05);(3)临床病理相关性分析表明mi R-638和Sox2的表达与肿瘤TNM分期(χ~2=10.617,P=0.001;χ~2=9.939,P=0.002)及门静脉侵犯与否明显相关(χ~2=7.885,P=0.005;χ~2=6.370,P=0.012),而与患者年龄、性别、肿瘤大小、肿瘤分化程度、肝炎状况等其他临床病理参数无关(均P>0.05)。结论肝癌中mi R-638表达水平的下调及SOX2表达水平的上升可能与肝癌的发生、发展密切相关。

Objective To investigate the expression of miR-638 and Sox2 and their clinicopathological significancein hepatocellular carcinoma （HCC）. Methods Carcinoma tissue and corresponding the adjacent normaltissue in 78 HCC patients were analyzed with qRT-PCR, to investigate the expression of miR-638 and Sox2. Immunohistochemistrywas also performed to detect the expression of Sox2 protein in 78 HCC patients. The correlationbetween the expression of miR-638 and Sox2 and the clinical features were statistically analyzed. Results（1）Compared with adjacent normal tissue, carcinoma tissue showed lower expressions of miR-638 levels, whileSox2 presented significantly higher expression （P〈0.05）. （2）Spearman rank correlation analysis showed that theexpression of miR-638 negatively correlated with the Sox2 protein expression in carcinoma tissue （r=-0.441,P〈0.05）. （3）Clinicopathological correlation analysis showed that reduction of miR-638 and elevation of Sox2 expressionswere marginally associated with tumor TNM stage （χ2=10.617, P=0.001, χ2=9.939, P=0.002） and portalvascular system invasion （χ2=7.885, P=0.005, χ2=6.370, P=0.012）. However, there was no significant differencein clinicopathological features, such as age, gender, tumor size, tumor differentiation, hepatitis status, etc （allP〉0.05）. Conclusion A down-regulation of miR-638 expression and up-regulation of Sox2 expression may beassociated with the occurrence and progression of HCC.
