摘要
为研究五指山猪SLA-DQA和SLA-DQB基因,获得基因序列并分析基因结构和相关遗传变异,试验采用PCR扩增猪SLA-DQA和SLA-DQB基因组序列,通过DNA测序寻找基因中的SNP位点。结果显示:在猪SLA-DQA基因组中共发现6处突变位点,分别位于该基因的启动子区、第1内含子、第2内含子、第3内含子和第5外显子中;在猪SLA-DQB基因组中共发现4处突变位点,分别位于该基因第1外显子和第5外显子中。本研究成功获得五指山猪SLA-DQA和SLADQB基因序列信息,为下一步五指山猪SLA-DQA和SLA-DQB基因的功能研究奠定基础。
This study aimed to detect the SLA-DQA and SLA-DQB genes, analyze their structures, and revealthe distribution of SNP in Wuzhishan pig and Yorkshire pig. SLA-DQA and SLA-DQB genes were amplified byPCR. Then the gene mutation was identified by sequencing. The results showed that six mutations were detectedin the SLA-DQA genomic fragment, one in the promoter region, two in the first intron, one in the second intron,one in the third intron and one in the fifth exon. Four mutations were detected in the SLA-DQB genomic fragment, three in the first exon and one in the fifth exon. In this study, we obtained the sequences of SLA-DQAand SLA-DQB genes which laid a foundation for further studying their functions in Wuzhishan pig.
