五味子多糖影响脑肿瘤干细胞的增殖 被引量:10

摘要 目的探讨五味子多糖对脑肿瘤干细胞增殖的作用。方法培养人脑肿瘤细胞,采用CD133免疫磁珠分选法获得脑肿瘤干细胞,MTT法检测五味子多糖对脑肿瘤干细胞生长增殖率的影响,ELISA法检测脑肿瘤干细胞分泌HIF、VEGF、Neun和GFAP蛋白变化。结果五味子多糖可以浓度依赖性地抑制脑肿瘤干细胞的增殖率,与对照组比较,800 mg/L组差异有统计学意义(P<0.01)。ELISA结果显示,五味子多糖作用后脑肿瘤干细胞分泌HIF和VEGF蛋白降低,分泌Neun和GFAP蛋白增高,与对照组比较,800 mg/L组差异有统计学意义(P<0.01)。结论五味子多糖通过诱导脑肿瘤干细胞分化和调控VEGF表达而抑制脑肿瘤干细胞的增殖。 Objective To analysis the effect of Schisandra chinensis polysaccharide(SCP) on the growth of brain tumor stem cells(BTSCs). Methods Human glioma cells were cultured,then BTSCs were obtained by CD133 immunomagnetic sorting assay. Expression of HIF,VEGF,Neun and GFAP proteins were examined by ELISA assay. Results Different doses of SCP could inhibit the proliferation rates of BTSCs,compared with control group,800 mg/L group was statistically significant(P 0. 01). Results of ELISA showed that,expressions of HIF and VEGF were decreased but Neun and GFAP were increased,compared with control group,800 mg/L group was statistically significant(P 0. 01). Conclusions SCP could induce the differentiation,and regulate VEGF protein,finally cause the growth inhibiting of BTSCs.
