老年男性代谢综合征患者外周血巨噬细胞中脂肪细胞型脂肪酸结合蛋白4表达水平及意义 被引量:2

Expression of adipocyte fatty acid-binding protein 4 in macrophages of elderly male patients with metabolic syndrome

摘要 目的探讨老年男性代谢综合征(MS)患者外周血单核源性巨噬细胞中脂肪细胞型脂肪酸结合蛋白4(FABP4)表达水平及其临床意义。方法选取2013年1月至2013年10月在南京医科大学附属常州第二人民医院就诊的老年男性患者108例。依据是否患MS分为MS组(n=68)和非MS组(n=40)。用酶联免疫吸附试验(ELISA)检测白细胞介素-6(IL-6)、肿瘤坏死因子α(TNF-α)、内皮生长因子(EGF)和血管内皮生长因子(VEGF);采用免疫比浊法检测高敏C反应蛋白(hs-CRP)水平,密度梯度离心法加贴壁法分离外周血中单核细胞,进行细胞培养,诱导分化成巨噬细胞,采用蛋白质印迹法检测巨噬细胞内FABP4表达水平。依据数据类型,分别采用t检验或x^2检验。指标间相关性采用Pearson相关分析。结果与非MS组比较,MS组患者的IL-6[(105.7±17.1)vs(135.4±11.8)pg/ml]、TNF-α[(20.60±6.19)vs(29.50±5.74)pg/ml]、hs-CRP[(1.98±1.00)vs(3.54±1.12)mg/L]、VEGF[(130.8±19.9)vs(180.2±48.1)pg/ml]及外周血巨噬细胞中FABP4表达水平[(0.25±0.13)vs(0.36±0.10)]均显著增高,差异具有统计学意义(P<0.05)。相关性分析表明,FABP4表达水平与VEGF(r=0.56)、IL-6(r=0.369)、TNF-α(r=0.441)、hs-CRP(r=0.597)均呈显著正相关(均P<0.05)。结论老年男性MS患者外周血单核源性巨噬细胞中FABP4表达水平升高,FABP4可能参与了MS患者血管内皮病变的发生发展,作用机制可能与炎症反应及血管内皮功能紊乱有关。 Objective To investigate the expression of adipocyte fatty acid binding protein(FABP4)in the macrophages of elderly male patients with metabolic syndrome(MS).Methods A total of 108 elderly patients who were admitted in our hospital from January to October 2013 were enrolled in this study.According to being diagnosed as MS or not,they were divided into MS group(n=68)and non-MS group(n=40).Serum levels of interleukin-6(IL-6),tumor necrosis factor α(TNF-α),epidermal growth factor(EGF)and vascular endothelial growth factor(VEGF)were detected by enzyme-linked immunosorbent assay(ELISA).And serum level of high-sensitivity C-reactive protein(hs-CRP)was measured by immunoturbidimetric assay.After the peripheral blood monocytes were obtained by Ficoll density gradient centrifugation and adherence method, the monocytes were differentiated to macrophages.The expression of FABP4 in the macrophages was determined by Western blotting.SPSS statistics 16.0 was used to analyze the data. According to data type,Students' t test or Chi-square test were adopted.Correlation between indices was analyzed by Pearson linear correlation analysis.Results Compared with the non-MS group,the serum levels of IL-6[(105.7 ±17.1)vs(135.4 ±11.8)pg/ml], TNF-α[(20.60 ±6.19)vs(29.50 ±5.74)pg/ml],hs-CRP[(1.98 ±1.00)vs(3.54 ±1.12)mg/L]and VEGF[(130.8 ±19.9) vs(180.2 ±48.1)pg/ml],as well as the expression of FABP4[(0.25 ±0.13)vs(0.36 ±0.10)]in the macrophages were signifi-cantly higher in the MS group(P 〈0.05).Correlation analysis showed that the FABP4 was positively correlated with VEGF (r=0.536),IL-6(r=0.369), TNF-α(r=0.441), and hs-CRP(r=0.597, all P〈0.05).Conclusion The protein level of FABP4 in the peripheral blood macrophages is high in the elderly male MS patients.FABP4 may be involved in the incidence and development of vascular endothelial impairments in MS patients, and the mechanism may be related to the inflammatory reaction and vascular endothelial dysfunction.
