摘要
超灵敏检测白细胞介素-8(IL-8)有助于口腔癌的早期诊断。本实验采用多孔铂取代天然的辣根过氧化物酶,催化双氧水(H2O2)实现信号转化与放大,构建了一种超灵敏的IL-8无酶电化学免疫传感器。采用透射电镜(TEM)和紫外可见吸收光谱(UV-Vis)对合成的多孔铂纳米酶进行形态和性能验证,利用交流阻抗法(EIS)、循环伏安法(CV)和方波伏安法(SWV)进行电化学性能表征。在最优的条件下,该方法的检测范围是100~5000fg/mL,检出限为85fg/mL(S/N=3)。将IL-8标准加入唾液中进行检测仍得到很好的结果,该方法对辅助口腔癌的早期诊断和治疗有潜在的应用价值。
We constructed a novel electrochemical human interleukin-8 (IL-8) immunosensor based on mesoporous platinum nanosphere (MPN) which is an non-enzymatic nanocatalyst. MPN plays an important role on the reduction of H2O2, and largely improves the sensitivity of the sensor. Transmission electron microscope (TEM) and UV-Vis absorption spectrometry were employed to characterize properties of nanomaterials. Electrochemical impedance spectroscopy (EIS), cyclic voltammetry (CV) and square wave voltammetry (SWV) were used to study the electrochemical properties of the sensor. Under optimum conditions, the amperometric signals increased linearly with the target IL-8 concentrations from 100 fg/mLto 5000 fg/mL, and the DNA sensor exhibited a detection limit as low as 85 fg/mL (S/N=3). It was also with a favorable signal response in saliva samples, which would become a simple and powerful tool to assist the early diagnosis and treatment of oral cancer.
