盐酸千金藤碱对鼻咽癌模型大鼠中IL-1β、IL-18、IL-12以及ICAM-1表达影响

Effect of Cepharanthine Hydrochloride on the Expressions of IL-1β,IL-18,IL-12 and ICAM-1 in Model Rats with Nasopharyngeal Carcinoma

目的:观察盐酸千金藤碱对鼻咽癌模型大鼠中IL-1β、IL-18、IL-12以及ICAM-1的表达影响。方法:将42只健康SD大鼠分为正常对照组、模型对照组、盐酸千金藤碱组各14只,正常对照组正常饲养,模型对照组以及盐酸千金藤碱组采用诱癌剂腋部皮下注射法以建立鼻咽癌的动物模型,给予诱癌剂二亚硝基哌嗪(DNP)15 mg/kg腋部皮下注射,每3 d注射1次,连续注射28次,同时给予促癌剂佛波醇酯(TPA)100 mg/kg,每周测量大鼠体质量以调整药物用量。盐酸千金藤碱组于模型建立后给予盐酸千金藤碱15 g·kg^(-1)·d^(-1)灌药,每日1次;其余两组给予蒸馏水灌药,剂量与使用频率和盐酸千金藤碱组相同至实验结束,各组均治疗3个月。比较各组大鼠情况、外周血和鼻腔冲洗液的IL-1β及IL-18、癌症组织中IL-12以及ICAM-1的表达情况。结果:实验过程中正常对照组无大鼠死亡,模型对照组大鼠8只死亡,死亡发生率为57.14%;盐酸千金藤碱组1只死亡,死亡发生率为7.14%。治疗后,与正常对照组相比,盐酸千金藤碱组、模型对照组白介素12平均灰度值较低(P<0.05),外周血和鼻腔冲洗液的白介素1β以及白介素18水平、细胞间黏附分子平均灰度值较高(P<0.05);与模型对照组相比,盐酸千金藤碱组白介素12平均灰度值较高(P<0.05),外周血和鼻腔冲洗液的白介素1β以及白介素18水平、细胞间黏附分子平均灰度值较低(P<0.05)。结论:盐酸千金藤碱具有良好的治疗鼻咽癌的效果,可能与下调癌症组织大鼠中ICAM-1的表达、外周血和鼻腔冲洗液的IL-1β及IL-18,上调IL-12表达有关,有望为临床治疗方面提供新的干预方法。

Objective：To observe the effect of cepharanthine hydrochloride on the expressions of IL-1 β , IL-18, IL-12 and ICAM-1 in model rats with nasopharyngeal carcinoma. Methods ： 42 health SD rats were divided into the normal control group, model control group, cepharanthine hydrochloride group, with 14 rats in each group. The rats in the normal control group were with normal feeding, the rats in model control group and eepharanthine hydrochloride group were cancer inducing agent subcutaneous injection to establish animal models with nasopharyngeal carcinoma, were given cancer inducing agent n-two （ DNP ） 15 mg/kg subcutaneous injection of axillary, injection each 3 days, a total of injection 28 times, at the same time gave tumor promoter phorbol ester alcohol （ TPA ） 100 mg/kg, measured the rats body quality weekly to adjust the amount of drugs. Cepharanthine hydrochloride group after established the models gave cepharanthine hydrochloride 15 g · kg-1 · d-1 irrigation medicine, once a day; The rest two groups were gaven distilled water irrigation medicine, and the dose and used frequency as the same as the cepharanthine hydrochloride group until theexperiment was over, each group treatment with three months. The rats situations, peripheral blood and nasal flush liquidof IL- 1 β ,IL-18, cancer tissues of IL-12 and ICAM-1 expressions situations were compared in every group. Results ： In the process of the experiment, the normal control group without rats died, the model control group with 8 rats died, the death rate was 57.14%; cepharanthine hydrochloride group with 1 rat died, the death rate was 7.14%. After treatment, compared with the normal control group, the interleukin-12 average gray value in the cepharanthine hydrochloride group and model control group were lower （ P〈0.05 ）. The peripheral blood and nasal flush liquid of interleukin -1β and interleukin-18 levels, the average gray value of intercellular adhesion molecule were higher （ P〈0.05 ）. Compared with the model control group, the interleukin-12 average gray value in the cepharanthine hydrochloride group was higher （ P〈0.05 ）, the peripheral blood and nasal flush liquid of interleukin -1 β and interleukin- 18 levels, the average gray value of intereellular adhesion molecule were lower （ P〈0.05 ）. Conclusion： Cepharanthine hydrochloride has a good effect in the treatment of nasopharyngeal carcinoma, and may be relate to reduce cancer tissues of ICAM -1 expression, peripheralblood and nasal flush liquid of IL-1 β, IL-18, increase IL-12, is expect to provide a new intervention method in clinical treatment.
