摘要
目的:系统研究影响大孔吸附树脂吸附分离的因素,筛选黄芪总皂苷的最佳吸附树脂和最佳工艺。方法:根据树脂的吸附-洗脱性能、吸附动力学曲线,从8种树脂中筛选出最佳吸附树脂,以黄芪总皂苷提取率、纯度为综合评价指标进行工艺优化。结果:AB-8、XDA-5、XDA-1三种树脂具有较优的吸附-洗脱性能,并依据吸附动力学曲线,确定AB-8型为最佳吸附树脂。工艺优化结果为AB-8型树脂,精密吸取样品液0.876 mg·mL^(-1)(总皂苷与干树脂质量比为34.6 mg·g^(-1))100 mL上柱,蒸馏水洗至无还原糖反应后,依次用10 BV 0.2%~0.3% NaOH溶液、10%乙醇和70%乙醇进行洗脱,流速为1~3 mL·min^(-1),收集70%乙醇洗脱部分,所得产物中总皂苷提取率和纯度分别为47.79%、68.98%。结论:该工艺简便可行,可较好的精制黄芪总皂苷。
Objective : Systematic study the influencing factors of macroporous resin adsorption separation, screening of the best resin and optimum technology, to provide the basis for purification Astragalus total saponins. Methods : The best adsorption resin from 8 kinds of resin were determinated according to the resin adsorption-elution and adsorption kinetics curve, and technology was be optimized with Astragalus total saponins extraction efficiency and purity as a comprehensive evaluation index. Results : AB-8, XDA-5, XDA-1, the three kinds of resin has better adsorption-elution. On the basis of the adsorption kinetics curve, AB-8 resin was determined as the best adsorption resin. The results of optimal technology were as follows : Concentration of sample solution was 0.876 mg · mL- 1 by AB-8 resin adsorption (quality ratio of Astragalus total saponins to dry resin was 34.6 mg · g-1), and distilled water to wash without reducing sugar reactions, total saponins was eluted with 10 BV 0.2%-0.3% NaOH solution, 10 BV 10% ethanol solution and 10BV 70% ethanol solution in turn, flow rate was controlled between 1-3 mL · min-1, 70% ethanol solution elution parts was collected, the extraction efficiency and the purity of total saponins in product was 47.79% and 68.98% respectively. Conclusion : The technology was be a simple and reliable method to purify Astragalus total saponins.
