摘要
目的 :探讨柴龙解郁丹对焦虑模型大鼠行为学的影响及其作用机制。方法 :采用不确定性空瓶刺激法建立大鼠焦虑模型,给予柴龙解郁丹干预1个月并以西酞普兰做对照,观察柴龙解郁丹对焦虑模型大鼠焦虑行为及海马单胺类神经递质(5-HT、DA、NE)含量的影响,单胺类神经递质采用酶联免疫吸附方法(ELISA)检测。结果:柴龙解郁丹能够显著改善模型大鼠焦虑行为优于西酞普兰组(P<0.01),降低大鼠海马区5-HT、DA、NE的含量(P<0.05),但对5-HT的调节作用不及西酞普兰(P<0.01),对DA、NE含量的影响功同西酞普兰(P>0.05),降低单胺类神经递质未呈现出量效关系。结论 :柴龙解郁丹有显著的抗焦虑作用,可能与降低焦虑模型大鼠海马单胺类神经递质而发挥抗焦虑作用有关。
Objective :To investigate the effect and action mechanism of Chailong Jieyu Pill on the behavior of rat models with anxiety disorder. Methods :Rat models with anxiety disorder were established using uncertain empty bottle stimulation. To observe the effects of Chailong Jieyu Pill on anxious behaviour and monoamine neurotransmitter(5-HT,DA,NE)in hippocampus of anxiety model rats,1 months were treated with Chailong Jieyu Pill,and citalopram was used as control. Enzyme linked immunosorbent assay was utilized to detect monoamine neurotransmitter. Results :Chailong Jieyu Pill can significantly improve the anxiety behavior in rats better than the citalopram group(P0.01),reduce the content of the rat hippocampus 5-HT,DA,NE(P0.05),but the regulatory effect of Chailong Jieyu Pill on 5-hydroxytryptamine was less than that of citalopram(P0.01). The effect on the content of DA and NE were the same as that of citalopram(P0.05).The decrease of monoamine neurotransmitter showed no dose effect relationship. Conclusions :Chailong Jieyu Pill has significant anti-anxiety effect probably through diminishing monoamine neurotransmitters in the hippocampus with rat models of anxiety disorder.
