Optimization of Ultrasonic Extraction Process of Total Triterpenoids from Poria Cocos (Schw.) Wolf by Response Surface Methodology

摘要 目的 :优选茯苓总三萜的超声波提取工艺。方法 :在单因素考察的基础上,以茯苓中总三萜的得率为评价指标,采用响应面分析法(选取乙酸乙酯为溶剂,料液比、提取温度、提取时间)确定最佳的超声波辅助提取工艺。结果 :最佳工艺为30倍体积的乙酸乙酯,超声时间32 min,超声温度35℃,茯苓总三萜的得率为0.685%。验证试验结果为0.683%,与预测值接近。结论 :Box-Behnken-设计的响应面优化茯苓总三萜的提取工艺方法简单,合理,可预测性优。 Objective :To optimize the ultrasonic-assist extraction process of total triterpenoids from Poria cocos. Methods :Response surface methodology was applied to optimize the best extraction conditions with the yield of total triterpenoids as index,on the basic of single factor tests,including liquid-solid ratio,extracting temperature and time. Results :The optimal conditions as follows :solidliquid ratio at 30∶1,the time for 32 min,extracting temperature at 35 ℃. The extraction rate of total triterpenoids from Poria cocos was 0.685%. The result of verification test was 0.683%,which was closed to the predicted value. Conclusion :The process is simple,reasonable and predictable,which can be used as the extraction process of triterpenes in P. cocos.
