摘要
目的 :研究XCT-790对沉默Dickkopf-1(Dkk1)腺病毒转染MG-63细胞活性和OPN、Lrp5、BMP2的影响。方法 :构建沉默Dkk1腺病毒载体,实验分为空白对照组、沉默Dkk1组、XCT-790处理空载腺病毒组、XCT-790处理沉默Dkk1组,根据组别不同分别用上述重组腺病毒转染MG63细胞,观察细胞增殖及OPN、Lrp5、BMP2表达情况。结果 :(1)与空白对照组比较,沉默Dkk1组和XCT-790处理空载腺病毒组MG63细胞活性增强,OPN、Lrp5、BMP2的表达量升高((P<0.05);(2)与Dkk1沉默组比较,XCT-790处理Dkk1沉默组的MG63细胞活性减弱,OPN、Lrp5、BMP2的表达量降低(P<0.05)。结论 :XCT-790可以降低因DKK1沉默而增强的MG63细胞活性,可以降低因DKK1沉默而增高的OPN、Lrp5、BMP2的表达量。
Objective :To investigate the effect of XCT-790 on the activity and OPN,Lrp5,BMP2 of MG63 cells infected with silence Dickkopf-1(Dkk1)adenovirus vector. Methods :Constructed recombinant adenovirus vector of silencing about Dkk1,the experiment was divided into control group,silence Dkk1 group,XCT-790 dispose no-load adenovirus group,XCT-790 dispose silence Dkk1 group,use these recombinant adenovirus to infect MG63 cells,then observe the cell proliferation and expression of OPN,Lrp5,BMP2. Results :(1)Compared with the control group,the activity and the expression of BMP2,Lrp5,OPN of MG63 cells in silence Dkk1 group and XCT-790 dispose no-load adenovirus group was increased(P0.05);(2)Compared with the silence Dkk1 group,the MG63 cell activity and expression of BMP2,Lrp5,OPN of XCT-790 dispose silence Dkk1 group both reduced(P0.05). Conclusion :XCT-790 can reduce effect of the higher MG63 cell activity and expression of BMP2,Lrp5,OPN that due to the silence Dkk1.
