摘要
目的 :通过基因芯片技术观察咳喘六味合剂对寒哮型支气管哮喘患者外周血表达谱影响并对相关目标基因进行实验验证。方法 :选取上海中医药大学附属龙华医院呼吸科门诊就诊的轻度寒性哮喘急性发作期患者64例,随机分为两组:咳喘六味合剂组(32例)和安慰剂对照组(32例)。两组患者分别口服咳喘六味合剂及安慰剂,20 mL/次,1 d 3次,疗程共1周。取患者干预后外周血行基因芯片检测,通过荧光实时定量PCR技术对相关基因进行结果验证。结果 :在表达谱中筛选出差异倍数1.5倍以上的基因233条。在选取的目标基因中,咳喘六味合剂组IL-8 mRNA、PDGFR-βmRNA的表达降低;IL-10 mRNA的表达升高,与基因芯片结果趋势一致,从而验证了基因芯片结果的准确性。结论 :咳喘六味合剂可能通过干预与免疫、炎症、气道重塑等相关基因的表达治疗寒性哮喘。
Objective :To observe the effects of Kechuan Liuwei Mixture on gene expression profile of peripheral blood in patients suffering asthma of cold type by gene chip detection and to prove the results by q RT-PCR. Methods :64 outpatients suffering asthma of cold type in Longhua hospital Shanghai university of traditional Chinese medicine were divided into Kechuan Liuwei Mixture group and placebo group,32 cases in each group. The two groups were asked to take Kechuan Liuwei Mixture and placebo 20 mL each time,three times a day,totally a week. Peripheral blood was sampled to perform gene chip detection. The results of some related genes were verified by a RT-PCR technology. Results :233 1.5-fold DEGs were screend from the gene expression spectrum. Among the target genes,IL-8 mRNA,PDGFR-βmRNA expressed down-regulated,while IL-10 mRNA expressed up-regulated. The results were consistent to the gene chip detection. Conclusion :The mechanism of Kechuan Liuwei Mixture treating asthma of cold type may relate to effecting genes that related to immune response,inflammatory reaction,airway remodeling.
