摘要
目的:分析板蓝根药材中3种核苷(尿苷、鸟苷、腺苷)高效液相法测定的含量。方法:用高效液相法测定板蓝根中尿苷、鸟苷、腺苷的含量,保持柱温在30℃,检测波长为254nm,梯度洗脱,流速保持在每分钟0.8m L。结果:尿苷在10.6~265.0 mg/L范围内线性关系良好,鸟苷在10.4~260.0 mg/L范围内线性关系良好。腺苷在10.8~270.0 mg/L范围内线性关系良好。平均回收率尿苷为101.2%,鸟苷为100.8%,腺苷为101.4%。测定精密度、重视性以及稳定性全部良好。结论:高效液相法测定板蓝根药材中3种核苷快速并且灵敏,检测方法简单,重视性理想,能够用于板蓝根药材的质量控制。
Objective: To analysis three kinds of nucleosides in Radix Isatidis ( uridine, guanosine, adenosine ) determined by HPLC method. Method: The content of uridine, guanosine and adenosine in Radix lsatidis was determined by HPLC, with keeping the column temperature in 30 degrees Celsius, the detection wavelength 254nm, gradient elution and the flow rate at 0.8ml per minute. Result: Uridine showed a good linear relationship at a range of 10.6 mg/L to 265.0 mg/L. Guanosine showed a good linear relationship at a range of 10.4 mg/L to 260.0 mg/L. Adenosine showed a good linear relationship at a range of 10.8 mg/ L to 270.0 mg/L. The average recovery rate was 101.2% in uridine, 100.8% in guanosine and 101.4% in adenosine respectively. Precision of determination, reproducibility and stability are all good. Conclusion: for the three nuclear glycosides in Radix Isatidis and HPLC method is a fast determination method, with sensitive detection, simple operation mode, good reproducibility in determination of Radix Isatidis, which could be used for the quality control of Radix Isatidis.
