摘要
目的:观察香青兰总黄酮对D-半乳糖/亚硝酸钠诱导的痴呆小鼠学习记忆的影响。方法:皮下连续注射D-半乳糖(120 mg·kg-1)/亚硝酸钠(90 mg·kg-1)60 d建立痴呆小鼠模型,观察受试药(25,50和100 mg·kg-1)对痴呆小鼠学习记忆的影响及对脑组织中SOD,MDA,GSH-PX,IL-1β,TNF-α和i NOS含量的影响。结果:与模型组相比,香青兰总黄酮对痴呆小鼠学习记忆有明显的改善作用;其中剂量组可显著降低MDA和IL-1β水平;高剂量组可明显提高SOD活性,下调MDA,IL-1β和TNF-α表达。结论:香青兰总黄酮可显著改善痴呆小鼠的学习、记忆能力,其机制可能与其降低脑组织中氧化水平、下调炎症介质的表达有关。
Objective: To observe the effects of total flavonoids of Dracocephalummoldavica L. on D-galactose/sodium nitrite-induced learning and memory disorders in mice. Methods: Sodium nitrite and D-galactose were subcutaneously injected for 60 days to induce dementia model in mice. The effect of total flavonoids of Dracocephalummoldavica L(25,50 and 100 mg·kg-1) on the learning and memory disorders in mice were evaluated by Morris water maze,and the levels of superoxide dismutase(SOD),malondialdehyde(MDA),glutathione peroxidase(GSH-Px),interleukin-1β(IL-1β),tumor necrosis factor-α(TNF-α) and nitric oxide synthase(i NOS) in the cortex were measured. Results: Compared with the model group,treatment with total flavonoids from Dracocephalummoldavica L. significantly improved the learning and memory deficits induced by D-galactose/sodium nitrite; the dose of 50 mg·kg-1 decreased the expression of MDA and IL-1β; the dose of 100 mg·kg-1 could obviously down-regulate the activity of SOD,and up-regulated the expressions of MDA,IL-1β and TNF-α. Conclusion: Total flavonoids from Dracocephalummoldavica L. can improve the ability of learning and memory in dementia mice and the mechanism is probably related to resisting oxidation damage and inflammatory reaction.
