多波长HPLC法同时测定桑菊银翘散中6种成分的含量 被引量:2

Simultaneous Determination of Six Constituents in Sang Ju Yinqiao Powder by HPLC under Multiple UV Wavelengths

摘要 目的建立多波长HPLC法同时测定桑菊银翘散中3,5-O-二咖啡酰基奎宁酸、绿原酸、连翘苷、连翘酯苷A、牛蒡苷和苦杏仁苷的含量。方法采用岛津公司的InertsilODS-SP C18柱(4.6×250mm,5μm);流动相为0.1%磷酸-乙腈,梯度洗脱;体积流量1.0m L·min-1;检测波长分别为苦杏仁苷210nm,连翘苷、牛蒡苷277nm,3,5-O-二咖啡酰基奎宁酸、绿原酸、连翘酯苷A 327nm;柱温35℃。结果 3,5-O-二咖啡酰基奎宁酸、绿原酸、连翘苷、连翘酯苷A、牛蒡苷和苦杏仁苷的线性范围分别为0.048~1.2μg、0.1024~2.56μg、0.0283~0.7075μg、0.0900~2.25μg、0.3802~9.505μg、0.0803~2.0075μg(r≧0.9995)。6种成分的平均回收率在97.1%~99.6%之间。结论该方法简便、准确、灵敏、重复性好,可用于测定桑菊银翘散中6种成分的含量。 OBJECTIVE To establish a multiple UV wavelengths HPLC method for simultaneously determina- tion the contents of 3,5-O-dicaffeoylquinic acid, Chtorogenic acid, Forsythin, Forsythoside A, Arctiin, and Amygdalin in SangJu Yinqiao powder. METHODS The analysis was conducted on the island ferry company Inertsil ODS-SP C18 column (4.6 × 250mm ,5μm), with 0. 1% (v/v) phosphoric acid-acetonitrile in a gradient elution mode as the mobile phase at a flow rate 1.0mL· min-1. The detection wavelength were set at 210nm for Amygdalin,277nm for Arctiin and Forsythin, and 327nm for 3,5-O-dicaffeoylquinic acid, Chlorogenic acid, Forsythoside A. The temperature was maintained at 35~C. RESULTS The linear ranges of 3,5-O-dicaffeoylquinic acid, Chlorogenic acid, Forsythin, Forsythoside A, Arctiin, and Amygdalin were 0. 048 - 1.2~g, 0. 1024 ~ 2. 56μg, 0. 0283 ~ 0. 7075μg, 0. 0900 2. 25μg,0. 3802 ~ 9. 505 μg ,0. 0803 ~ 2. 0075μg ( r ≥0. 9995 ), respectively. The average recoveries were within the ranges of 97.1% -99.6%. CONCLUSION The developed method is simple, specific, aceurate and with good re- producibility, which can be used for quality control of SangJu Yinqiao powder.
