摘要
目的 研究整合素αvβ3单克隆抗体抗人胆管癌血管生成的作用。 方法 将人胆管癌细胞系QBC939细胞接种于鸡胚尿囊膜 (CAM)无血管区 ,给予整和素αvβ3单克隆抗体 ,观察其对人胆管癌血管生成及肿瘤生长的影响。结果 整合素αvβ3单克隆抗体组血管数目在给予抗体后第 2~ 8天均显著低于磷酸盐缓冲液 (PBS)对照组 (P <0 .0 5) ,光镜下瘤细胞分布稀疏 ,瘤组织新生血管腔减少。结论 整合素αvβ3单克隆抗体能够抑制胆管癌新生血管的生成 ,具有显著的抗肿瘤作用。
Objective To investigate the inhibitory effects of monoclonal antibody against integrin αvβ3 on angiogenesis induced by human bile duct carcinoma.Methods Cells from human bile duct carcinoma cell line QBC939 were transplanted to the non vascular region of the chick embryo chorioallantic membrane, and the monoclonal antibody of integrin αvβ3 was given, its effects on angiogenesis and tumor growth were examined.Results The number of the newly formed vessels upon the tumor was significantly decreased in the group using monoclonal antibody against integrin αvβ3 as compared with the PBS control group on days 2 to 8 after the treatment (P<0.05 ). Optical microscopy showed the decrease in the tumor population and the newly formed vessels.Conclusion The monoclonal antibody against integrin αvβ3 can inhibit the angiogenesis induced by human bile duct carcinoma and has a remarkable anti tumor effect.
