摘要 采用EDC法制备获得黄曲霉毒素B1人工抗原AFB1-BSA。对AFB1-BSA紫外扫描,结果表明其图谱与黄曲霉毒素B1和BSA的图谱有明显差异,其红外扫描结果与BSA的图谱也有明显差异,表明BSA与黄曲霉毒素B1偶联成功。以不同摩尔比的AFB1-BSA人工抗原免疫小鼠,制备了特异性的鼠抗AFB1多克隆抗血清。综合评价不同起始摩尔比反应所得人工抗原的免疫特性和AFB1转化率,当AFB1与BSA的起始摩尔比为40∶1时,AFB1与BSA的偶联产物摩尔比为4.93∶1,AFB1的转化率达12.33%;用间接ELISA法测其抗血清的效价,经过四次免疫之后,抗血清的效价最高,高达1∶1 056 000。 Aflatoxin B1 artificial antigen (AFB1 -BSA)was prepared with EDC method. The results showed that the UV spectrum of AFB1-BSA was different from BSA and Aflatoxin B1 obviously, and the IV spectrum was also different from BSA distinctly, all of which indicated that the preparation of AFB1-BSA was successful. BALB/c mice were immunized wth different molar ratio of AFB1 -BSA artificial antigen, which made preparation of the specific polyclonal antiserum against AFB1. This paper comprehensively evaluated the immune characteristics of artificial antigens of different initial molar ratio and the utilization rate of AFB1. The artificial antigen was obtained with the molar ratio of 4.93 : 1 and the AFB1 utilization rate of 12.33%, when the initial molar ratio of Aflatoxin B1 to C-BSA was 40 : 1. BALB/c mice were immunized with AFB1-BSA for 4 times, and their titer antiserum from those mice were up to 1 : 1 056 000 testified by indirect ELISA method.
